二甲丸对大鼠增生型子宫内膜雌、孕激素受体及bcl-2、bax表达的影响

Effect of Erjia pill on estrogen receptor and progesterone receptor and expression of bcl-2 and bax in hyperplasia of endometrium in rats

目的观察益肾软坚中药二甲丸对大鼠增生型子宫内膜中雌、孕激素受体及凋亡调节蛋白bcl-2和bax表达的影响,探讨其治疗子宫内膜增生的作用机制。方法将80只健康雌性SD大鼠随机分为空白组、模型组、中药高剂量组、中药中剂量组、中药低剂量组及西药组,除空白组外,其余组均行去势术,术后第3天采用补佳乐灌胃建立大鼠子宫内膜增生模型。造模成功2 d后,中药高、中、低剂量组分别给予二甲丸常用量的2倍、常用量、常用量的0.5倍灌胃,西药组给予地屈孕酮灌胃,空白组和模型组给予等量蒸馏水灌胃,均连续14 d。采用免疫组化SP法检测各组大鼠子宫内膜中雌激素受体(ER)、孕激素受体(PR)、bcl-2和bax的表达水平。结果中药高剂量组ER、bcl-2均明显低于模型组(P均<0.05),PR、bax均明显高于模型组(P均<0.05)。相关性分析显示,bax与ER表达呈负相关,bcl-2与ER表达呈正相关;bax与PR表达呈正相关,bcl-2与PR表达呈负相关。结论子宫内膜过度增生大鼠ER和bcl-2表达增加,PR和bax表达下降。益肾软坚中药二甲丸通过影响ER和PR表达水平来调节bax以及bcl-2的表达以促进子宫内膜细胞凋亡,从而改善子宫内膜增生状态。

Objective It is to observe the effect of Erjia pill on estrogen receptor（ ER） and progesterone receptor（ PR）and expression of apoptosis regulatory protein bcl-2 and bax in hyperplasia of endometrium in rats,and to explore its action mechanism in the treatment of endometrial hyperplasia. Methods 80 healthy female SD rats were randomly divided into blank group,model group,high,medium and low dose groups,western medicine group,all the rats except in blank group were given castrated surgery,on the 3th postoperative the rats were given Bujiale by intragastric infusion to establish models of endometrial hyperplasia. In 2 days after model successfully establishing,high,medium and low dose group were given Erjia pills at 2time normal dosage,normal dosage and 0. 5 time dosage respectively by intragastric infusion,the western medicine group was given flexor progesterone,blank group and model group was given the same dosage of distilled water,all the rats were treated for 14 d. ER and PR and expression of bcl-2 and bax in hyperplasia of endometrium was detected by immunohistochemical SP method in each group. Results The levels of ER and bcl-2 in high dose group were significantly lower while that of PR and bax were higher than those in model group（ P all〈 0. 05）. Correlation analysis showed that there was negative correlation between bax and ER,bcl-2 and PR,positive correlation between bcl-2 and ER,bax and PR. Conclusion Yishen Ruanjian TCM Erjia pill can promote the apoptosis of endometrial hyperplasia cells by influencing ER and PR expression to regulate the expression of bax and bcl-2,and thus to improve the state of endometrial hyperplasia.
