摘要 目的优化栀子多酚超声提取工艺。方法通过Plackett-Burman试验从乙醇浓度、粒径、液料比、超声功率、超声温度、超声时间、提取次数几个影响因素中挑选出显著因素,然后针对显著因素采用最速上升试验确定响应面的中心点,最终利用Box-Behnken试验优化栀子多酚超声提取工艺。结果最佳提取工艺为乙醇浓度45%,液料比25:1,提取时间35min,栀子总多酚得率为1.857%,与预测值1.9%接近。结论优化工艺科学合理,为栀子的开发提供了参考。 Objective To optimize the ultrasonic extraction process of polyphenols from Gardenia jasminoides Ellis. Methods The significant factors were selected from the influence factors of concentration of ethanol, particle diameter、the ratio of liquid to material, ultrasonic power, ultrasonic temperature, ultrasonic time and extraction times by Placket-Burman experiment, then the central point of response surface was determined by the Steepest ascent experiment, finally the ultrasonic extraction process was optimized by Box-Behnken experiment. Results Optimum ultrasonic extraction process was as follows: concentration of ethanol 45%, the ratio of liquid to material 25:1, ultrasonic time 35 min, the total yield of polyphenol was 1.857%, close to the predicted value of 1.9%. Conclusion The optimized extraction technology which provided references for the exploitation and utilization of Gardenia jasminoides Ellis was scientific and reasonable.
