摘要
采用高效液相色谱法测定乌饭树茎中槲皮素的含量.通过单因素实验和正交实验优化槲皮素的提取工艺条件.结果表明,回流法提取乌饭树茎中槲皮素最佳条件是:以甲醇:25%HCl(4∶1,V/V)做提取剂,料液比1∶60(g/m L),浸泡时间7 h,回流时间70 min,水浴温度70℃.在此工艺条件下槲皮素提取率为0.079%.
Process parameters of quercetin extraction from Vaccinium bracteatun stems by reflux method was studied. Quercetin content in Vaccinium bracteatun stems was determined by high performance liq- uid chromatography (HPLC) method. The optimal process of quercetin extraction was determined by single factor experiment and orthogonal experiment. The results showed that the rate of extraction quer- cetin from Vaccinium bracteatun stems reached 0.079% under the best extraction conditions of metha- nol - 25% HC1 (4: 1, V/V) as extraction agent, 1 : 60 (g/mL) material liquid ratio, 7 h immersion time, 70 minutes extraction by reflux method and 70℃water bath temperature.
