高盐饮食对大鼠血浆AngⅡ、CGRP、ICAM-1、VCAM-1和P-S的表达的影响 被引量:4

摘要 目的探讨高盐饮食对大鼠血压及血浆AngⅡ、CGRP、ICAM-1、VCAM-1和P-S的表达的影响.方法选取8周龄雄性Wistar大鼠80只,随机分为对照组(30只)和高盐组(50只),分别给予正常饮水和2%Na Cl高盐饮水,喂养21 d,每周测量大鼠尾动脉收缩压.实验结束时,采用酶联免疫吸附实验(enzyme linked immunosorbent assay,ELISA)检测大鼠血浆血管紧张素Ⅱ(angiotensionⅡ,AngⅡ)、降钙素基因相关肽(calcitonin gene-related peptide,CGRP)、细胞间粘附分子-1(intercellular adhesion molecule-1,ICAM-1)、血管间粘附分子-1(vascular adhesion molecule-1,VCAM-1)、P选择素(P-selectin production,P-S)的水平.结果与对照组相比,高盐组大鼠实验7 d后血压既有显著升高,且高血压状态一直持续到实验结束(P<0.05).实验21 d后,高盐组大鼠血浆AngⅡ、ICAM-1、VCAM-1和P-S水平显著高于对照组(P<0.05),相关性分析显示AngⅡ、ICAM-1、VCAM-1和P-S水平与血压均正相关(r=0.813,P<0.01;r=0.641,P<0.05;r=0.693,P<0.05;r=0.703,P<0.05);高盐组血浆CGRP水平低于对照组(P<0.05),且血浆CGRP水平与血压负相关(r=-0.683;P<0.05).结论高盐饮食可影响大鼠血管内皮舒张功能以及引起血管炎症反应. Objective To investigate the effect of highsalt diet on blood pressure and expression of AngⅡ,CGRP, ICAM1,VCAM1 and PS in plasma of rat.Methods 80 Wistar rats(8 weeks,male) were randomly divided into control group(fed with tap water,n=30) and highsalt group(fed with highsalt water contraining 2% Na Cl,n=50).The blood pressure was measured every seven days.After being fed for 21 days,the levels of AngⅡ,CGRP,VCAM1,ICAM1 and PS in plasma were detected by ELISA.Results After 7 days of highsalt dietary, the systolic blood pressure in highsalt group was significantly higher than that of the control group(P〈0.05).The levels of AngⅡ,ICAM1,VCAM1 and PS in highsalt group were significantly higher than those of the control group(P〈0.05),and the levels of AngⅡ,ICAM1,VCAM1 and PS were found to be positively correlated with blood pressure(r=0.813,P〈0.01;r=0.641,P〈0.05;r=0.693,P〈0.05;r=0.703,P〈0.05);The level of CGRP in highsalt group was significantly lower than that of the control group(P〈0.05), and was negative correlated with blood pressure(r =0.683; P〈0.05).Conclusions High salt diet could significantly impair the function of vascular endothelial diastolic and cause vascular inflammation.
