Slit2-Tg小鼠行为异常的病理表型

摘要 目的观察Slit2-Tg转基因小鼠类似神经退行性变行为的病理表型特征。方法利用Morris水迷宫及避暗实验装置检测Slit2-Tg小鼠的学习记忆能力,HE染色观察Slit2-Tg小鼠大脑皮层与海马神经元病理改变情况,免疫组化检测小鼠脑组织皮层和海马神经元核抗原、乙酰胆碱酯酶(Ach E)、β-淀粉样蛋白(Aβ)的表达情况,Western blot检测小鼠脑组织Aβ、Ach E的表达水平,并与野生型C57BL/6J小鼠比较。结果与野生型C57BL/6J小鼠比较,Slit2-Tg小鼠行为出现异常,表现记忆力明显下降;病理学观察发现皮层与海马区有明显的神经元损伤,神经元空泡变性,数目减少;同时发现皮层和海马区毒性蛋白Aβ的沉积增多(P<0.05),Ach E的活性明显增强(P<0.01)。结论 Slit2-Tg小鼠脑部有明显的损伤,并出现类神经退行性变的病理学特点,对进一步研究Slit2蛋白过表达对脑神经系统的影响提供指导意义。
