某氡温泉周边居民外周血淋巴细胞中miRNAs的表达 被引量:3

摘要 目的:探讨河北省某地氡温泉对周围居民外周血淋巴细胞中miR-16、miR-106b、miR-449a、miR-34a和let-7g表达的影响。方法:采用简单随机抽样法抽取某地氡温泉周边居民43人;同时随机抽取生活习惯相似,但未长期接触过氡温泉的居民44人。采用实时荧光定量PCR(qPCR)方法检测研究对象淋巴细胞中miRNAs的表达水平。用t检验比较miRNA在氡温泉组和对照组居民外周血淋巴细胞中的差异表达;采用多重线性回归分析miRNA的差异性表达和居民氡暴露的关系。结果:与对照组相比,氡温泉组居民外周血淋巴细胞中miR-106b、miR-449a和let-7g表达水平显著上调(t值分别为4.180、2.422、2.794,P均<0.05)。控制年龄、性别、吸烟和饮酒等因素后,miR-106b、miR-449a和let-7g的差异性表达和居民氡暴露因素呈正相关(t值分别为4.091、2.716、2.440,P均<0.05)。但是miR-16和miR-34a的表达改变与氡暴露无显著相关(t值分别为0.990、0.147,P均>0.05)。结论:miR-106b、miR-449a和let-7g具有作为氡致辐射损伤早期标志物的潜在价值。 OBJECTIVE:To investigate the expressions of miR-16,miR-106 b,miR-449 a,miR-34 a and let-7g in peripheral blood lymphocytes of the residents around hot springs with radon in Hebei province.METHODS:Two groups were randomly selected in Hebei province,Radon group consisted of 43 residents around hot springs with radon and control group were 44 residents with similar living conditions but not around hot springs.qRT-PCR was used to detect miRNAs level in radon and control groups,t test was used for statistical analysis of miRNA levels in lymphocytes between two groups.Multiple linear regression analyse were employed to test the association between miRNAs expression and radon exposure.RESULTS:Compared with control group,the levels of miR-106 b,miR-449 a and let-7g in lymphocytes of radon group were significantly increased(t=4.180,2.422,2.794;P〈0.05).After controlling of confounding factors such as age,gender,smoking and drinking,alteration of miR-106 b,miR-449 a and let-7g in lymphocytes were positively correlated with radon exposure(t=4.091,2.716,2.440;P〈0.05).No significant difference was observed in the lymphocyte levels of miR-16 and miR-34 a between the two groups(t=0.990,0.147;P〈0.05).CONCLUSION:MiR-106 b,miR-449 a and let-7g levels could potentially used as biomarkers for radon exposure.
