摘要 泛素-蛋白酶体系统(UPS)是真核细胞内特异性蛋白降解系统,参与多种生物学活动,包括细胞周期调控、信号转导、细胞凋亡、蛋白质转录及翻译等。UPS异常与人类许多恶性肿瘤、神经退行性病变、心血管疾病及糖尿病等疾病的发生、发展有关。UPS各组分已成为许多疾病的研究焦点。UPS与线粒体关系密切,不但参与线粒体蛋白质量控制、维持线粒体形态,而且直接介导线粒体自噬。 The ubiquitin-proteasome system(UPS) is a specific route of protein degradation in eukaryotic cells. Moreover,it has become clear that the UPS fulfills an important function in most aspects of cukaryotic biology, such as regulation of cell cycle, intracellular signaling, transcriptional control and apoptosis. Any abnormality in UPS may lead to illnesses such as malignancies, neurodegenerative disorders, cardiovascular disease and diabetes. UPS components have become the research focus of many diseases. Recent studies dem- onstrated a close relationship between UPS and mitochondrion. Accumulating evidence shows that the UFS plays an essential role not only in mitochondrial protein quality controls and dynamics, but also in the initiation of mitochondrial mitophagy.
