白细胞介素-4诱导人支气管上皮细胞ORMDL3基因表达的时效性 被引量:1

Time-effect of interleukin 4 on inducing the expression of ORMDL3 gene in human bronchial epithelial cells

摘要 目的:观察白细胞介素-4(IL-4)诱导人支气管上皮细胞(16HBE)不同时间血清类黏蛋白1样蛋白3(ORMDL3)的表达,探讨在细胞模型中IL-4对ORMDL3的转录和合成的影响。方法:用100ng/m L的IL-4诱导支气管上皮细胞后,分别在不同时间段(0、12、18、24、36、48h)用RT-PCR法检测ORMDL3 m RNA的表达,Western Blot法检测ORMDL3蛋白的表达。结果:正常支气管上皮细胞有微量的ORMDL3表达,IL-4诱导细胞后,ORMDL3m RNA和蛋白的表达均增加,在一定范围内呈时间依懒性,与对照组比较差异有统计学意义(P<0.05),其他各组与24h组比较差异有统计学意义(P<0.01)。结论:IL-4能诱导ORMDL3高表达,并且在一定范围内呈时间依赖性。 Objective: To observe the expression of ORMDL3 in human bronchial epithelial cells(16 HBE) induced by interleukin 4(IL-4) at different time and explore the effect of IL-4 on the transcription and synthesis of ORMDL3 in cell model. Methods: IL-4(100ng/m L) was used to induce the bronchial epithelial cells. At 0, 12, 18, 24, 36 and 48 h after experiment, RT-PCR was used to determine the expression of ORMDL3 m RNA, and Western Blot was used to determine the expression of ORMDL3 protein. Results: There was few ORMDL3 in the normal bronchial epithelial cells, however, the expressions of ORMDL3 m RNA and protein were both increased by IL-4 that depended on the time in a certain extent. Furthermore, there was a statistical difference in the expression of ORMDL3 between the experimental groups and the control group(P〈0.05). While, in all experimental groups, there were significant statistical differences in the expression of ORMDL3 between the 24 h experimental group and the others(P〈0.01). Conclusion: IL-4 could induce an overexpression of ORMDL3, which had a time-effect relationship in a certain extent.
