基于PGF_(2α)浓度变化研究当归川芎对抗原发性痛经小鼠子宫组织痉挛的机制 被引量:21

摘要 目的从PGF2α浓度变化出发,研究当归-川芎对原发性痛经小鼠子宫组织痉挛的作用及其机制;方法采用雌二醇联合缩宫素建立原发性痛经(primary dysmenorrhea,PD)模型,将当归:川芎不同比例(1∶1、2∶1、3∶1、3∶2)作用于PD小鼠,观察小鼠扭体反应潜伏期及30min内扭体次数,用ELISA法测定小鼠子宫组织中前列腺素F2α(PGF2α)、前列腺素E2(PGE2)、孕酮(P)浓度变化;结果当归-川芎4种配比均能提高扭体潜伏期,减少扭体次数(P<0.05、0.01);均能降低PGF2α的浓度(P<0.01),提高PGE2及P的水平(P<0.05、0.01);PGF2α浓度与扭体次数呈正相关(P<0.01),与扭体反应潜伏期呈负相关(P<0.05);PGE2、P与扭体次数及扭体潜伏期之间无相关性(P>0.05);PGF2α与PGE2、P均呈现出负相关(P<0.05、0.01);结论当归-川芎能对抗PD小鼠子宫平滑肌痉挛,其机制与抑制PGF2α生成,提高PGE2、P水平相关。 Objective: To study the function and mechanism of Angelica- Ligusticum wallichii for uterine tissue spasm of primary dysmenorrhea mice on the base of PGF concentration changes. Methods : Primary dysmenorrhea (PD) mouse model was established by using estradiol and oxytocin. Angelica -Ligusticum wallichii were put in different proportions (1: 1,2: 1, 3: 1, 3:2) in PD mice to observe writhing response latency and writhing times within 30 minutes and to measure prostaglandin F2α ( PGF2α ) , prostaglandin E2 ( PGE2 ) , progesterone (P) concentration of the mouse uterine tissue by ELLSA. Result: 4 kinds of ratio of Angelica - Ligusticum walliehii could increase writhing latency and reduce writhing times ( P 〈 0. 05, 0. 01 ) and also reduce the concentration of PGF2= ( P 〈 0. 01 ) and increase the level of PGE2 and P ( P 〈 0. 05, 0. 01 ). PGF2α concentration was positively correlated with the writhing times ( P 〈 0.01 ) and negative- ly correlated with the writhing response latency ( P 〈 0. 05 ). PGE2 and P had no eorrelation with writhing times and latency ( P 〉 0. 05 ). PGF2α showed a negative correlation with PGE2 and P (P 〈 0. 05, 0. 01 ). Conclusion: Angelica - Ligusticum wallichii can fight against uterine smooth muscle spasm of PD mice and its mechanism and inhibition of PGF2α production are correlated with an increase of PGEz and P levels.
