复方大黄软膏中黄芩苷的含量测定及体外透皮特性 被引量:3

摘要 的:建立复方大黄软膏中黄芩苷的含量测定方法,并考察其体外透皮特性。方法:采用HPLC法测定黄芩苷含量,色谱柱为Agilenn200-C18柱(4.6mm×250mm,5μm),流动相为甲醇-水-磷酸(47:53:0.2),流速为1.0mL/min,检测波长为280nm;应用药物透皮扩散试验法考察其体外透皮特性。结果:黄芩苷的质量浓度在1.5172~242.75μg/L范围内与峰面积呈良好线性关系(r=1),平均加样回收率102.1%(RSD%=2.7%,n=6);2批次复方大黄软膏中黄芩苷的含量分别为2.31mg/g、2.17mg/g,经离体裸小鼠皮肤24h平均累积透过率仅为0.25%。结论:该方法简便、快速、准确,可用于复方大黄软膏的质量控制及透皮特性评价。 Objective: To establish the method for the content determination of baiealin in compound rhubarb ointment, and to investigate its percutaneous permeation characteristic in vitro. Methods: The content of baicalin was determined by HPLC. The determination was performed on Agilent1200-C18 (4.6 mm×250 mm,5 μm) column with mobile phase consisted of methanol- water- phosphoric acid(47:53:0.2) at the flow rate of 1.0mL/min. The detection wavelength was set at 280 nm. The pereutaneous permeation characteristic in vitro was observed by the method of drug transdermal diffusion test. Results: The linear range of baicalin was 1.5172-242.75 μg/mL (r=l) with an average recovery of 102.1%(RSD%=2.7%,n=6). The average content of baicalin in two batches compound rhubarb ointment were 2.31 mg/g and 2.17 rag/g, respectively. The average cumulative penetration rate of baicalin in 24 h was only 0.25%. Conclusion: The method is proved to be simple, rapid, and accurate, and can be used for the quality control of compound rhubarb ointment and percutaneous permeation characteristics research.
