摘要 固定化金属螯合亲和层析介质,因其具有固定化配基选择简单且稳定性高、复杂环境中一步纯化蛋白、特异性吸附、目标蛋白质吸附量大、蛋白洗脱条件温和、介质易再生、重复利用率高、制备工艺简单、造价低等多项优点,逐渐成为蛋白质分离纯化及多个相关领域中最有效的分离技术。通过综述固定化金属螯合亲和层析介质的工作原理、构成及应用现状,对固定化金属螯合亲和层析介质应用的前景进行展望。 Immobilized Metal-chelated Affinity Chromatography (IMAC) has gradually became the most effective technology in purifying protein and multi-correlation sphere, due to the choice of ligand is simple and high steady, single step purification protein in complex environment, specific adsorption, great amount of target protein adsorption, mild elution condition, renewable, high utilization rate, preparation technology is simple, low-cost and so on. The principle, constitution, application of IMAC are introduced in this paper, as well as the prospect of IMAC.
