金荞麦叶发酵茶抗小鼠H22肿瘤的作用及机制研究 被引量:18

摘要 为探究金荞麦叶发酵茶抑制肿瘤的作用及其机制,以移植H22荷瘤小鼠为模型,设置空白组、金荞麦叶发酵茶低中高剂量组(1.25、2.50和5.00 g·kg-1·d-1)灌胃,阳性药5-FU组,检测其抑瘤效果及对H22小鼠生存时间的影响。以荷瘤小鼠血清中SOD和MDA含量检测评价金荞麦叶发酵茶的抗氧化能力,以耳肿胀度、鸡红细胞(CRBC)致敏小鼠血清溶血素水平、碳廓清能力及小鼠免疫器官的重量评价免疫功能的调节能力。结果发现,金荞麦叶发酵茶高剂量能够明显抑制小鼠H22实体瘤生长(P<0.05),其抑瘤率为36.3%;SOD活力与模型组比较显著提高(P<0.05),而低剂量组、中剂量组和高剂量组MDA含量较模型组均下降,但与模型组比无明显差异。发酵茶能显著提高正常小鼠DTH反应所致的耳廓肿胀度(P<0.01);可提高CRBC致敏小鼠的血清溶血素含量,且显著加强小鼠单核巨噬细胞的吞噬能力,加快碳粒的清除,而对正常小鼠免疫器官的影响比较复杂。可见,金荞麦叶发酵茶通过显著增强小鼠的免疫功能,提高SOD的活性,清除自由基,减轻活性氧带来的损伤,从而起到抗肿瘤作用。 In order to research the anti-tumor function and the mechanism of gold buckwheat fermented tea, a group of H22 tumor-transplanted mice were cultivated by gavage of gold buckwheat fermented tea at the dose of 1.25, 2.50 and 5.00 g.kg-1.d-1 representing low, medium, and high dose groups, respectively, and normal mice were used as control. The positive control group was established by gavage of 5-FU in mice with H22 tumor. The tumor-inhibition effect and the survival time of mice with H22 tumor were detected. SOD and MDA concentrations in mice serum were used to evaluate the antioxidant activity of gold buckwheat fermented tea. In addition, The ear swelling degree, serum hemolysin level of hypersensitive mice induced by CRBC and carbon particle clearance and weight of immune organs were used to evaluate the ability of regulating immune function of gold buckwheat fermented tea. As results, the high-dose gold buckwheat fermented tea could notably inhibit the growth of H22 tumor in mice (P〈0.05) with an inhibitory rate of 36.3 %. Besides, SOD activity increased clearly compared with the model group (P〈0.05). The MDA concentration in the low, medium, and high dose groups decreased compared with the model group; however, the decreased effect was not significant. The gold buckwheat fermented tea could notably improve the ear swelling degree incurred by DTH response in normal mice (P 〈0.01). It also elevated the serum hemolysin level of CRBC sensitized mice and significantly enhanced the ability of mononuclear phagocytic macrophages in mice, as well as accelerated the clearance of carbon particles. The impact of the gold buckwheat fermented tea on immune organs in mice is complicated. In conclusion, the gold buckwheat fermented tea can deploy antineoplastic function in mice by enhancing the immune function, augmenting SOD activity, clearing free radicals and al- leviating lesions generated by reactive oxygen.
