人参皂苷Rb1、维生素B_(12)抑制亚砷酸钠诱导人脐静脉内皮细胞凋亡及其机制的研究

摘要 目的:探讨Na As O2诱导HUVECs细胞凋亡前提下人参皂苷Rb1、维生素B_(12)单独或共同作用对HUVECs细胞的保护作用,为地砷病的防治提供新途径。方法 :将HUVECs细胞随机分为对照组、Na As O2组、Rb1+Na As O2组、维生素B_(12)+Na As O2组、Rb1+维生素B_(12)+Na As O2组;采用Annexin V-FITC/PI双染检测细胞凋亡;用DCFH-DA染料检测细胞内活性氧(ROS)水平。结果 :人参皂苷Rb1、维生素B_(12)单独及共同作用均可明显保护Na As O2诱导的HUVECs凋亡;同时发现细胞内ROS水平也显著下降,但二者在降低ROS方面无相加作用。结论:人参皂苷Rb1、维生素B_(12)单独应用均能有效保护Na As O2诱导的内皮细胞凋亡,其作用机制可能是通过降低细胞内ROS水平实现。 Objective:To investigate the protective effect of Ginsenosides Rbl and Vitamin B12 alone or in combination on apoptosis of human umbilical vein endothelial cells (HUVECs)induced by sodium arsenite. nethods.HUVECs cells were cultured and randomly divided into blank control group,sodium arsenite group, Ginsenosides Rbl+sodium arsenite group,Vitamin B12+sodium arsenite group and Ginsenosides Rbl+Vitamin B12+sodium arsenite group.Apoptosis was detected by flow cytometry with Annexin V-FITC/PI dual staining;the levels of intracellular reactive oxidative species(ROS)were measured by flow cytometry with a cell based ROS assay.Results:Ginsenosides Rbl and Vitamin B12 alone or in combination inhibited apoptosis of HUVECs in- duced by sodium arsenite;they inhibited ROS generation in HUVECs induced by sodium arsenite without ad- ditive effect.Conhision:Ginsenosides Rbl and Vitamin B12 inhibited the apoptosis of HUVECs induced by sodium arsenite,which was associated with the decrease of intracellular levels of ROS generation.
