摘要
目的:探索中药纳达合剂对胆汁反流性胃炎(BRG)大鼠胃黏膜及相关炎症因子的影响。方法:采用灌胃牛胆酸钠+胰酶+卵磷脂法制备胆汁反流性胃炎模型。正常组、模型组灌胃生理盐水,纳达合剂低、中、高剂量组及麦滋林组分别予灌胃相应药物干预。实验结束,通过病理切片HE染色观察各组大鼠胃黏膜炎症变化情况,ELISA法检测大鼠胃黏膜白介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)、细胞间黏附分子-1(ICAM-1),Western blot方法检测大鼠胃黏膜单核细胞趋化因子-1(MCP-1)。结果:模型组大鼠胃黏膜炎症细胞浸润明显,炎症积分较正常组显著增高(P<0.01);与模型组比较,各药物干预组均可以显著改善大鼠胃黏膜炎症情况(P<0.01),其中纳达合剂组呈剂量依赖性,且与麦滋林组比较差异无统计学意义(P>0.05)。模型组大鼠胃黏膜炎症因子TNF-α、IL-8、ICAM、MCP-1蛋白表达较正常组显著增高(P<0.01);与模型组比较,纳达合剂各剂量组呈剂量依赖性地显著降低TNF-α、IL-8、ICAM、MCP-1蛋白表达(P<0.05或P<0.01),药麦滋林组亦可显著减少胃黏膜TNF-α、IL-8、ICAM、MCP-1蛋白表达(P<0.01),而纳达中、高剂量组在改善TNF-α表达方面优于麦滋林组(P<0.05或P<0.01),纳达高剂量组在改善ICAM-1表达方面优于麦滋林组(P<0.01)。结论:纳达合剂可显著改善胆汁反流性胃炎大鼠的胃黏膜病理,并对相关炎症因子有明显的抑制作用。
Objective: To observe the effects of"Na Da Mixture"on TNF-α,IL-8,ICAM-1 and MCP-1 in gastric mucosa of rat bile reflux gastritis. Methods: Preparation of bile reflux gastritis model by intragastric administration of sodium taurocholate,trypsin and lecithin. Normal group and model group were perfused with physiological saline,Na Da mixture of low,middle,high dose groups and Marzulene group were given intragastric administration of the corresponding drug for intervention.Finally,HE staining was used to observe the inflammation extent of gastric mucosa. ELISA test was used to detect the content of TNF-α,ICAM-1,IL-8 in gastric mucosa tissue. Western blot analysis was used to detect the expression of MCP-1.Results: Model group had a significant rise of inflammatory cells infiltration and inflammation score compared with control group( P 〈0. 01). Compared with model group,each drug intervention group could significantly improve pathological changes of gastric mucosa( P〈 0. 01),in which"Na Da Mixture"of each dose decreased the inflammation score in a dose-dependent mannerand had no statistical differencecompared with Marzulene group( P 〈0. 05). Model group had a significant rise of TNF-α,ICAM-1,IL-8,MCP-1 compared with control group( P 〈0. 01). Compared with model group,"Na Da Mixture"of each dose could significantly decrease the content of TNF-α,ICAM-1,IL-8 and MCP-1 in gastric mucosa tissue in a dose-dependent manner( P 〈0. 05 or P 〈0. 01). Marzulene also had the similar effect( P〈 0. 01). Compared with Marzulene group,"Na Da Mixture"of middle and high dose had a better improvement in TNF-α expression( P 〈0. 05 or P 〈0. 01). Meanwhile,"Na Da Mixture"of high dose had a better improvement in ICAM-1 expression( P 〈0. 01). Conclusion: "Na Da Mixture "can significantly improve pathological changes of gastric mucosal of BRG rats,and obviously suppress associated inflammatory factors.
