摘要 目的研究环氧合酶2(cyclooxygenase 2,COX-2)对大鼠血管平滑肌细胞(vascular smooth muscle cells,VSMCs)增殖、迁移和表型转化的影响。方法体外培养大鼠采集VSMCs,采用噻唑蓝(Methyl thiazolyl tetrazolium,MTT)法、Transwell小室法观察不同浓度COX-2对VSMCs增殖、迁移的影响;RNA干扰技术沉默COX-2后,观察其对VSMCs增殖、迁移的影响;Western bot法分析COX-2-si RNA对VSMCs表型标志蛋白的影响。结果 COX-2可促进VSMCs增殖、迁移,并呈现浓度依赖关系;COX-2-si RNA则可显著抑制VSMCs的增殖、迁移;Western blot法结果表明,COX-2-si RNA可上调VSMCs收缩型标志蛋白的表达,同时下调VSMCs合成型标志蛋白的表达。结论 COX-2可以促进VSMCs的增殖和迁移,同时促进其表型的转化。 Objectives To investigate the effects of cyclooxygenase 2(COX-2) on the proliferation, migration and phenotypic transformation of vascular smooth muscle cells(VSMCs) in rats. Methods Methyl thiazolyl tetrazolium(MTT) assay and Transwell were used to determine the effects of COX-2 on the proliferation and migration of VSMCs.COX-2 was then silenced by si RNA(small interference RNA) and the proliferation and migration were detected in VSMCs transfected with COX-2-si RNA. The expression of VSMC marker proteins were assayed by Western blot. Results COX-2 could significantly promote the proliferation and migration of VSMCs in a concentration-dependent manner.Meanwhile, COX-2-si RNA could obviously inhibit the proliferation and migration of VSMCs. In addition, COX-2-si RNA could up-regulate the expression of VSMC contractile marker protein and down-regulate the expression of VSMC synthetic marker protein. Conclusions COX-2 can promote the proliferation, migration and phenotypic transformation of VSMCs simultaneously.
