猪繁殖与呼吸综合征糖玻璃活疫苗制备及其耐热性能的初步研究 被引量:1

摘要 采用蔗糖、胰蛋白胨等成分配伍的耐热保护剂处方T28C-5,对猪繁殖与呼吸终审合征病毒(PRRSV)进行了真空梯度干燥,获得性状为干燥致密的白色泡沫状疫苗。采用扫描电镜对PRRS泡沫疫苗和传统的冻干商品疫苗进行了形貌分析,发现泡沫疫苗呈现糖玻璃样,而冻干疫苗呈现疏松孔道样。DSC测定结果表明,泡沫疫苗具有较高的Tg,为57.45℃。干燥的疫苗在37℃保存4个月,病毒效价损失≤1.0 Lg;25℃保存5个月,病毒滴度损失仍<0.8 Lg;自然环境温度下保存190 d(历尽夏季、秋季、冬季),病毒滴度损失仍<0.6 Lg。将37℃、25℃及4℃下保存3个月的泡沫干燥疫苗免疫20日龄仔猪,免后2周ELISA抗体水平合格且与商品苗趋势相当。试验结果表明泡沫干燥疫苗具有卓越的耐热性能。
