The influence of different conservation condition of EDTA-K_2 anticoagulant venous blood on leukocyte classification

摘要 目的观察EDTA-K2抗凝静脉血24 h内不同温度下保存对白细胞分类的影响。方法将EDTA-K2静脉血于5℃和常温下,用SE-9000血细胞分析仪作白细胞分类(仪器法),同时制作薄血片,经瑞氏染色后人工分类计数白细胞(手工法)。结果手工法正常中性粒细胞在常温8 h与0 h相比开始明显减少,差异有统计学意义(P<0.05);2种温度下10 h均显著减少,差异有统计学意义(P<0.01);正常单核细胞在2种温度下12 h开始明显减少,差异有统计学意义(P<0.05);淋巴细胞和嗜酸性粒细胞在24 h内的百分比差异均无统计学意义(P>0.05)。仪器法常温下5类白细胞在24 h内各时段的百分比差异均无统计学意义(P>0.05);5℃8 h单核细胞明显增加,24 h中性粒细胞增加、淋巴细胞和单核细胞减少,差异均有统计学意义(P<0.05)。结论静脉抗凝血采集后,应尽快做白细胞分类,特殊情况不能及时处理时也应在8 h内制片染色备用。 Objective To observe the influence of conserving the anticoagulant venous blood of different temperature during 24 hours on leukocyte classification. Methods EDTA - K2venous blood was put on 5 ℃ and common temperature separately, and differential leukocyte count were respectively detected by SE -9000 automatic hematology analyzer( instrumental method). Sim- uhaneously thin blood smear was made and stained by Wright' s staining to classify and count leukocyte by artificial ( hand - made method). Results Differential leukocyte count by hand - made methods, compared with 0 h, the percentage of the eu- morphism neutrophil at 8 h on common temperature reduced obviously, and the difference was statistically significant (P 〈 0.05 ), and that at 10 h on two temperatures reduced notably, and the difference was statistically significant (P 〈 0.01 ) ; the percentage of the eumorphism monocyte on two temperatures at 12 h decreased significantly, and the difference was statistically significant( P 〈 0.05 ) ; there was no statistical significance on the difference of the percentage of leukomonocyte and eosinophilic granulocyte at 24 h( P 〉0.05 ). Through the instrumental method, the difference on the percentages of five leucocyte on com- mon temperature had no statistical significance at each stage within 24 h ( P 〉 0.05 ) ; but for samples conserved at 5 ℃, the percentage of the mononuclear ceils increased obviously at 8 h, so did neutrophil and lymphoeytes and mononuclear decreased at 24 h(P 〈 0.05 ). Conclusion After collecting anticoagulant venous blood, leukocyte classification should be carried out quickly. If sample could not be detected immediately when in some special circumstances, it would be best to make thin blood smear and stain within 8 h.
