手术对卵巢癌患者外周血调节性T细胞的影响及临床意义 被引量:1

摘要 目的探讨手术前后卵巢癌患者外周血CD4+CD25+Foxp3+、CD8+CD28-及CD8+Foxp3+调节性T细胞的变化及其临床意义。方法流式细胞术检测31例卵巢癌患者术前及术后外周血CD4+CD25+Foxp3+、CD8+CD28-及CD8+Foxp3+调节性T细胞的表达水平,并与20例卵巢良性肿瘤及20例健康体检者做比较研究。结果卵巢癌患者外周血CD4+CD25+Foxp3+、CD8+CD28-及CD8+Foxp3+调节性T细胞的表达水平显著高于卵巢良性肿瘤及健康体检者(P<0.01),Ⅲ/Ⅳ期卵巢癌患者外周血调节性T细胞表达水平显著高于Ⅰ/Ⅱ期患者(P<0.05);卵巢癌患者外周血调节性T细胞的表达水平与病理类型无关;手术后卵巢癌患者外周血调节性T细胞的比例显著低于手术前(P<0.05);Ⅰ/Ⅱ期卵巢癌患者手术后调节性T细胞的比例与健康体检者相比无统计学差异,而Ⅲ/Ⅳ期病人术后调节性T细胞的比例仍显著高于健康体检者(P<0.05)。结论 CD4+CD25+Foxp3+、CD8+CD28-及CD8+Foxp3+调节性T细胞与卵巢癌的病理进程及肿瘤负荷相关。 This study performed to investigate the changes of CD4+CD25+Foxp3+, CD8+CD28-and CD8+Foxp3+regulatory T cells in peripheral blood of ovarian cancer patients and its clinical significance. The percentages of CD4+CD25+Foxp3+, CD8+CD28-and CD8+Foxp3+regulatory T cells in peripheral blood from 31 patients with ovarian cancer was detected by flow cytometry before and after operation, and compared with those in 20 patients with benign ovarian tumor and 20 healthy volunteers. Data showed that the percentages of CD4+CD25+Foxp3+, CD8+CD28-and CD8+Foxp3+Tregs in ovarian cancer patients were significantly higher those in 20 patients with benign ovarian tumor and 20 healthy volunteers. Levels of these Tregs in peripheral blood of patients at stage Ⅲ/Ⅳ was significantly higher than that of patients at stage Ⅰ/Ⅱ(P〈0.05); no differences of theses Tregs were found between serous ovarian cancer and mucinous ovarian cancer(P〈0.05). The levels of theses Tregs were dropped significantly after operation(P〈0.05). Furthermore, postoperative Treg percentages in stage Ⅰ/Ⅱ was not statistically different from that of healthy volunteers, while postoperative Treg percentages in stage Ⅲ/Ⅳ remained above the values shown by healthy volunteers(P〈0.05). In conclusion, the percentages of CD4+CD25+Foxp3+, CD8+CD28-and CD8+Foxp3+Tregs are correlated with the pathological stage and tumor burden in ovarian cancer.
