摘要
目的建立血液中的乙醇浓度的顶空气相色谱内标定量测定方法。方法采用DB-1大口径毛细管柱,以叔丁醇为内标,手动顶空气相色谱火焰离子化检测器进行测定,内标法、峰面积比定量。结果乙醇浓度在0.04~5.0 m/ml范围内,乙醇与内标物的峰面积比值与浓度呈线性关系(r=0.999 6),检出限为0.01 mg/ml,样品加标回收率为97.2%~107.8%,日内精密度RSD〈9.3%,日间精密度RSD〈5.9%。结论该法操作简便快速,干扰少,灵敏,准确,适用于血液中乙醇含量的检验。
[Objective] To establish a headspace gas chromatography method for determining blood alcohol concentration.[Methods]D-1 large diameter capillary column was adopted, tert butyl alcohol as internal standard, manual headspace gas chromatography flame ionization detector was used for detection, quantified on the basis of peak area ratios. [Results] A calibration curves were linear(r =0.999 6) between the ratio of the peak area and concentration of alcohol was obtained in the range of 0.04-5.0 mg/m L. The limit of detection was 0.01mg/ml,the recoveries were 97.2%-107.8% RSD within-day〈9.3%, RSD between-day 〈5.9%.[Conclusion] This method is simple, sensitive, and accurate with less interference and can be applied to determination of blood alcohol.
