脐带血CD34^+细胞使用ProCOUNT方法与双平台ISHAGE方法检测的比较分析

摘要 目的评估ProCOUNT方法和双平台ISHAGE方法检测脐带血CD34^+细胞计数的相关性及优缺点。方法采用流式细胞仪ProCOUNT和双平台ISHAGE对228份脐带血进行CD34^+细胞计数,对结果进行配对t检验和Spearman分析。结果 228份脐带血经ProCOUNT方法和双平台ISHAGE方法检测的CD34+绝对计数、CD34^+相对计数、有核细胞数差异均无统计学意义(P>0.05)。2种方法绝对计数具有强相关性(r=0.824,P<0.001),相对计数具有中相关性(r=0.714,P<0.001)。结论 2种方法可在检测CD34^+细胞时相互通用,为脐带血库的检测方案提供更多的选择。 Objective To evaluate the correlation,advantages and disadvantages of ProCOUNT and double-platform ISHAGE assays for detecting CD34^+ cells in cord blood samples. Methods CD34^+cells of 228 fresh cord bloods were detected using flow cytometric ProCOUNT and double-platform ISHAGE assays. The results of two methods were compared using Spearman correlation and paired t-test. Results There was no statistical significance on the differences of absolute number of CD34^+cells,the relative number of it,and the number of karyocyte( P〈0. 05). The absolute number of CD34^+ cells between two methods was highly correlated( r = 0. 824,P〈0. 001),and that of the relative number was moderate correlated( r = 0. 714,P〈0. 001). Conclusion Two methods can be substituted each other when the CD34^+ cells were detected. It provided more choice for umbilical cord blood bank.
