摘要
目的:研究中期因子在乳腺癌患者血清中的表达及其临床意义。方法:选择乳腺癌患者45例和乳腺良性肿瘤45例进行研究,收集乳腺肿瘤标本检测MK的mRNA含量、收集血清检测MK的蛋白含量;培养乳腺癌MCF-7细胞株,转染不同浓度的MK表达质粒后,检测细胞增殖和凋亡情况、培养基中VEGF的表达量以及细胞中MMPs、TIMPs的表达量。结果:乳腺癌患者乳腺组织中MK的表达量以及血清中MK的含量均高于乳腺良性患肿瘤患者,TNM III/IV期、低/未分化、淋巴结转移乳腺癌患者乳腺组织中MK的表达量以及血清中MK的含量均高于TNM I/II期、中/高分化、淋巴结未转移乳腺癌患者;MK表达质粒能够以剂量依赖性的方式增加乳腺癌细胞株中MK的mRNA含量和蛋白含量、增加细胞活力、降低细胞凋亡比例;100.0μg/mL质粒组培养基中VEGFA、VEGFB、VEGFC的含量以及细胞中MMP2、MMP9的含量明显高于0μg/mL质粒组,细胞中TIMP1、TIMP2的含量低于0μg/mL质粒组。结论:乳腺癌患者血清中中期因子含量异常升高,MK的高表达能够诱导乳腺癌细胞增殖、抑制乳腺癌细胞凋亡、促进血管新生过程和细胞侵袭过程。
Objective: To study serum midkine expression in breast cancer patients and its clinical significance. Meth- ods= 45 patients with breast cancer and 45 patients with benign breast tumor were selected for the study, breast tumor speci- mens were collected to deteet mRNA content of MK and serum was collected to detect protein eontent of MK; breast eancer MCF-7 cell lines were cultured and transfected with varying concentrations of MK expression plasmid, and then cell prolifera tion and apoptosis, VEGF expression in media as well as MMPs and TIMPs expression in cells was detected. Results: MK ex- pression in breast tissue and serum MK content of breast cancer patients were higher than those of benign breast tumor pa- tients, and MK expression in breast tissue and serum MK content of breast cancer patients with TNM Ⅲ/Ⅳ stage, low/un- differentiation and lymph node metastasis were higher than those of breast cancer patients with TNM Ⅰ/Ⅱ stage, medium/high differentiation and without lymph node metastasis; MK expression plasmid could dose-dependently increase mRNA content and protein content of MK in breast cancer cell lines, increase cell viability and decrease apoptosis percentage; VEGFA, VEGFB and VEGFC contents in media as well as MMP2 and MMP9 contents in cells of 100.0 μg/mL plasmid group were significantly higher than those of 0μg/mL plasmid group, and contents of TIMP1 and TIMP2 in cells were significantly lower than those of 0 μg/mL plasmid group. Conclusion.. Serum midkine content in breast cancer patients abnormally rises, and high expression of MK can induce breast cancer cell proliferation, inhibit breast cancer cell apoptosis and promote angiogenesis and cell invasion.
