摘要
目的:研究子痫前期胎盘中Gadd45α的表达量及敲低Gadd45α对滋养细胞HTR8/Svneo的调节作用。方法:收集子痫前期胎盘组织和正常胎盘组织,分别采用荧光定量PCR法和免疫印迹法检测Gadd45α的mRNA含量和蛋白含量;培养滋养细胞HTR8/Svneo,转染Gadd45α的siRNA后检测细胞的侵袭能力及侵袭相关分子的表达量。结果:子痫前期胎盘组织中Gadd45α的mRNA含量和蛋白含量高于对照组;转染Gadd45α的siRNA后,HTR8/Svneo细胞中Gadd45α的mRNA含量和蛋白含量均明显降低,侵袭细胞的数目以及MMP1、MMP2、MMP3、MMP9的表达量均明显增加。结论:子痫前期胎盘中Gadd45α的表达量异常升高;抑制滋养细胞HTR8/Svneo中Gadd45α的表达量能够促进的侵袭、增加MMPs分子的表达。
Objective: To study the Gadd45α expression in preeclampsia placenta and the regulation effect of Gadd45α on trophoblastHTR8/Svneo. Method: Preeclampsia placenta tissue and normal placental tissue were collected, mRNA content and protein content of Gadd45α were detected by real--time PCR and Western blotting. Trophoblast HTRS/Svneo were cultured and cell invasion ability and invasionrelated molecules were detected after transfection of Gadd45α siRNA. Results.. mRNA con- tent and protein content of Gadd45α in preeclampsia placenta mRNA content and protein content was higher than normal pla- cental tissue. After transfection of Gadd45αsiRNA, mRNA content and protein content of Gadd45α in HTR8 / Svneo cell were significantly reduced, the number of invasive cells and expression levels of MMP1, MMP2, MMP3, MMP9 were significantly increased. Conclusion: The expression of Gadd45α in preeclampsia placenta is abnormally increased. Suppressing expression of Gadd45α in trophoblast cells HTR8 / Svneo can promote cell invasion and increaseexpression of MMPs molecules.
