极低频电磁场对骨肉瘤HOS细胞的体外生物效应 被引量:2

摘要 目的:观察极低频电磁场(extremely low frequency electromagnetic field,ELF-EMF)对骨肉瘤HOS细胞的增殖、凋亡、克隆形成及侵袭能力的作用,筛选出对骨肉瘤HOS细胞抑制作用最佳的极低频电磁场参数。方法:体外培养骨肉瘤HOS细胞,分别固定电磁场频率为1Hz、5Hz、10Hz、15Hz、20Hz、30Hz、40Hz、50Hz,选择不同电磁场强度0.1mT、1mT、5mT、10mT、15mT,分组进行辐照,并设立对照组。采用CCK-8法检测细胞增殖情况并计算抑制率。经统计学分析后,筛选出2组对HOS细胞抑制作用较强的ELF-EMF参数,再通过检测其细胞凋亡、克隆形成及侵袭能力进行验证。结果:各组OD值经统计学分析,显示(1Hz,0.1mT)实验组细胞增殖抑制率最高,(40Hz,10mT)实验组次之(P<0.05)。流式细胞术结果表明,这2组的HOS细胞凋亡比率均高于对照组(P<0.05)。细胞克隆实验中,对照组克隆率最大,(1Hz,0.1mT)实验组次之,而(40Hz,10mT)实验组克隆率最小(P<0.05)。侵袭实验中,(40Hz,10mT)实验组穿过小室基底膜的细胞数量明显低于其他两组(P<0.05)。结论:(40Hz,10mT)组ELF-EMF参数对骨肉瘤HOS细胞有更明显抑制增殖作用,促使细胞凋亡,降低细胞的克隆能力,并减低其侵袭能力。 Objective:By the ELF - EMFg (extremely low frequency electromagnetic field) effects on the osteosarcoma HOS cells'proliferation, apoptosis, invasion and colony formation, the optimal inhibition parameter of ELF - EMF on osteosarcoma HOS cells could be filtered finely. Methods : HOS cells were irradiated with different magnetic field intensities(0.1 mT, 1 mT, 5mT, 10mT, 15mT) , which were fixed with magnetic field frequencies ( 1Hz, 5Hz, 10Hz, 15 Hz,20Hz,3OHz ,40Hz, 50Hz ) , comparing with the control groups, in vitro. HOS cells' proliferation and inhibition rate was calculated with CCK -8 assay. With the statistical analysis, we selected two stronger inhibition of ELF - EMF parameter on HOS cells, and validated them with the assay of apoptosis, colony formation and invasion again. Results : When the OD values of every group were analyzed,the cell inhibition rate of (1Hz ,0.1 mT) group of ELF -EMF irra- diation was the most,and (40Hz, 10mT) group was the second (P 〈 0.05 ). Both of the two groups'apoptosis rates were higher than the control group,with the flow cytometry (P 〈 0.05 ). In the colony formation assay, the most effi- ciency of clone formation was the control group,the second was (1Hz,0. hnT) group,and the least was (40Hz, 10mT) group(P 〈 0.05). In addition,the number of invased HOS cells in (40Hz, 10mT) group was obviously less than others ( P 〈 0.05 ). Conclusion: ( 40 Hz, 10mT) group of the ELF - EMF parameter can significantly inhibite the proliferation of osteosarcoma HOS cells, which can also induce apoptosis, reduce the ability to clone and slow down the invasion.
