环氧合酶-2抑制剂对非小细胞肺癌A549细胞Bcl-2和Bax表达的影响 被引量:2

Effect of cyclooxygenase-2 inhibitor on the expression of Bcl-2 and Bax in A549 cell lines of non-small cell lung cancers

摘要 目的探讨环氧合酶-2抑制剂对非小细胞肺癌A549细胞Bcl-2和Bax表达的影响。方法加不同浓-1度塞来昔布与A549肺癌细胞共培养,MTT法检测细胞增殖,50μmol·L塞来昔布作用于A549细胞48 h,实时定量PCR方法检测各组细胞的Bcl-2和Bax基因表达水平,Western blot方法检测各组细胞的Bcl-2和Bax蛋白表达。结果塞来昔布在体外对细胞株A549的生长抑制作用呈剂量、时间依赖性;各组细胞培养48h后,相比于对照组,塞来昔布组细胞Bcl-2 m RNA和蛋白表达显著下调,Bax m RNA和蛋白表达显著上调,Bcl-2/Bax比值显著降低(<0.01)。结论塞来昔布可能通过调节Bcl-2和Bax的表达抑制A549细胞增殖。 Objective To study the effect of cyclooxygenase-2 inhibitor on the expression of Bcl-2 and Bax in A549 cell lines of non-small cell lung cancers. Methods The proliferation of A549 lung carcinoma cells was measured by MTT assay after A549 cells were treated with different doses of celecoxib. After A549 cells were treated with celecoxib 50μmol·L-1 for 48 h, the expressions of Bcl-2 and Bax m RNA and protein were detected by real-time PCR and Western blot respectively. Results Celecoxib could inhibit the proliferation of A549 cells with a time-and dose-dependent manner. Compared with control group, the expression level of Bcl-2 was decreased and the expression level of Bax was increased significantly after A549 cells were treated with-1celecoxib 50μmol·L for 48 h, and the rate of Bcl-2/Bax was decreased significantly( P〈0.01). Conclusion Celecoxib inhibits the proliferation of A549 lung carcinoma cells by regulating the expressions of Bcl-2 and Bax.
