树舌灵芝发酵产漆酶培养基优化 被引量:5

摘要 在单因素试验的基础上,采用Plackett-Burman设计对影响树舌灵芝产漆酶活性的因素进行了评价,筛选出具有显著影响的因素并优化了树舌灵芝产漆酶培养基的主要成分。结果表明,产酶培养基中小麦麸皮、豆粕、硫酸铜和香兰素的添加量对树舌灵芝产漆酶活性具有显著影响;预测得到最佳培养基组成为:小麦麸皮(20 g/L)、豆粕(2 g/L),硫酸铜(0.625 g/L)和香兰素(0.037 5 g/L);优化培养基后漆酶活性达到45.85 IU/m L,约为优化前的56倍,为大规模发酵生产漆酶提供技术支持。 On the basis of single factor experiment, the Plackett-Burman experimental design was applied to evaluate the factors that potentially influence the laccase activity of Ganoderma applanatum. The most important factors were selected to optimize the culture medium. Results showed that, different levels of wheat bran, soybean meal, copper sulphate and vanillin could significantly influence the laccase activity. The most optimized culture medium included wheat bran(20 g/L), soybean meal(2 g/L), copper sulphate(0.625 g/L) and vanillin(0.037 5 g/L).Laccase activity increased up to 45.85 IU/m L, which was approximate 56 times before the optimization. This study provided the technical support for large scale fermentation of laccase.
