基于粪代谢组学的痛风脾虚证实质研究 被引量:16

Essential research on gout with spleen deficiency syndrome based on fecal metabonomics

摘要 目的:运用核磁共振氢谱(1H-NMR)方法研究脾虚型痛风患者粪便代谢标记物。方法:收集痛风患者和健康人群粪便,利用1H-NMR技术对各组粪便上清进行代谢谱检测。数据用SIMCA-P软件进行正交偏最小二乘-判别分析(OPLS-DA)和正交信号校正-偏最小二乘-判别分析(OSC-PLS-DA)挖掘代谢产物信息,并对差异代谢进行方差分析。结果:与健康人群相比,痛风患者粪便中丁酸、丙酸含量减少(P<0.05,P<0.01),乙醇、谷氨酰胺、牛磺酸、葡萄糖含量增加(P<0.05)。与非脾虚型痛风患者相比,脾虚型痛风患者粪便中异亮氨酸、亮氨酸含量增多(P<0.05),甘氨酸含量减少(P<0.05)。结论:建立了基于人粪便上清的1H-NMR代谢组学研究方法,获得了与痛风疾病及脾虚证型相关联的粪便代谢标志物,对痛风的预防和治疗具有一定的启发作用。 Objective: To investigate the fecal metabolic markers of gout patients with spleen deficiency syndrome based on 1H-NMR metabolomics. Methods: Analysis of fecal metabolic profiles in gout patients with syndrome of spleen deficiency and healthy individuals was carried out by 1H-NMR. The data was dealt with SIMCA-P through orthogonal partial least squares discriminant analysis(OPLS-DA) and orthogonal signal correction partial least-squares discriminant analysis(OSC-PLS-DA). One-way AVOVA was employed to analyze metabolites differences. Results: Compared with healthy individuals, the abundance of butyric acid and propionic acid of the gout patients decreased(P<0.05, P<0.01), while the content of ethanol, glutamine, taurine and glucose increased(P<0.05). Furthermore, the concentrations of isoleucine, leucine and glycine between spleen-deficiency and non-spleen-deficiency gout patients showed apparent difference(P<0.05). Conclusion: The 1H-NMR-based metabolomic approach coupled with multivariate statistical analysis for fecal metabolomic is established and corresponding metabolic markers associated with gout and spleen-deficiency syndrome are illuminated, facilitating the prevention and treatment of gout.
