石斛合剂通过GLP-1/β-Catenin信号通路促进胰岛细胞增殖的机制 被引量:9

Mechanism of Dendrobium Compound to promote proliferation of islet cell by GLP-1/β-Catenin signaling pathway

摘要 目的:探讨石斛合剂对胰岛β细胞保护作用及可能机制。方法:采用体外细胞培养的方法,予GLP-1受体抑制剂(Exendin9-39)干预胰岛β-TC-6细胞,MTT测细胞存活率,流式细胞仪测细胞周期,Western blot检测相关蛋白表达变化。结果:抑制剂干预后细胞S期缩短(P<0.01);石斛合剂组及阳性对照组S期延长(P<0.05,P<0.01)。抑制剂干预后细胞GLP-1R、β-Catenin、p-β-Catenin(ser-675)、PDX-1蛋白表达显著下调(P<0.01);石斛合剂组及阳性对照组GLP-1R、β-Catenin、p-β-Catenin(ser-675)、PDX-1蛋白表达水平明显上调(P<0.05,P<0.01)。结论:石斛合剂可保护胰岛β细胞,促进其增殖,其机制可能是通过增加GLP-1R受体及其相关蛋白的表达实现的。 Objective: To study the protective effect and possible mechanism of Dendrobium Compound(DC) on islet cell. Methods: The method of cell culture in vitro was adopted to handle islet cell by GLP-1R inhibitor(Exendin9-39). Then MTT was used to determine the cell survival, and cell cycle was measured by flow cytometry. Western blot was used to detect the related protein expression. Results: Cells' S phase was shortened after interference by inhibitor(P<0.01). The S phases in DC group and positive control group were prolonged(P<0.05, P<0.01). The related protein expressions of GLP-1R, β-Catenin, p-β-catenin(ser-675), PDX-1 after interference by inhibitor were significantly reduced(P<0.01). The protein expression levels of GLP-1R, β-Catenin, p-β-catenin(ser-675), PDX-1 in DC group and positive control group were significantly increased(P<0.05, P<0.01). Conclusion: Dendrobium Compound can protect islet β cell, and promote its proliferation, whose mechanism might be achieved by increasing the expression of GLP-1R receptor and its related protein.
