摘要
目的:检测新扶正除疫颗粒对肺炎链球菌所致小鼠急性上呼吸道感染模型肺组织中TLR2、TLR4mRNA变化的影响,探究新扶正除疫颗粒在抗炎及免疫方面的作用机制。方法:小鼠分为空白组、模型组、新扶正除疫颗粒的高中低剂量组、中药对照组和西药对照组,造模1h后灌药,并于造模后第1、4、7天取材,采用Real-time PCR方法测定小鼠肺组织中TLR2、TLR4 mRNA。结果:小鼠肺组织中TLR2在第1、4天各药物干预组与空白组和模型组比较,差异显著(P<0.05),第7天各组与模型组差异显著,但除低剂量组外均与空白组无显著性差异。肺组织中TLR4在第1、4天时,高、中剂量组、中药对照组及西药对照组与模型组有差异,但与空白组无显著差异。各组均存在随给药时间TLR2、TLR4 mRNA含量出现下降的趋势,新扶正除疫颗粒高、中剂量组优于低剂量组,与西药对照组无显著性差异。结论:提示新扶正除疫颗粒可以激活TLR2/TLR4系统,从而达到抗炎与免疫调节的作用,增强免疫与抗炎抑菌作用和中医"扶正"与"祛邪"的治法有关联性。
Objective: To determine the effect of new Fuzheng Chuyi Granule on TLR2 and TLR4 mRNA in mice pulmonary tissue infected with streptococcus pneumoniae, and to investigate the mechanism underlying new Fuzheng Chuyi Granule anti-inflammatory and regulating immunity function. Methods: The mice were divided into control group, model group, high, middle and low dosage groups of new Fuzheng Chuyi Granule, TCM control group and western medicine control group. The mice were intragastric administration after one-hour modelling, and sacrificed at 1th day, 4th day and 7th day after modelling, and then the TLR2 and TLR4 mRNA in mice pulmonary tissue were detected by Real-time PCR. Results: There was significant difference in the expression of TLR2 mRNA in mice pulmonary tissue between drug intervention groups and either blank group or model group on the first and fourth day(P<0.05). Similarly, there was difference between drug intervention groups and model group on the seventh day, when no significant difference was found between control group and drug intervention groups except for low dosage group. There was significant difference in TLR4 in pulmonary tissue among high dosage group, western medicine and TCM control groups and modeling group, but there was no significant difference when compared to blank group. There was a descending tendency in the expression of TLR2 and TLR4 mRNA depending on the drug administration time in every group. Additionally, high and middle dosage groups of new Fuzheng Chuyi Granule were superior to the low dosage group, which was found no significant difference when compared with western medicine group. Conclusion: New Fuzheng Chuyi Granule can activate TLR2/TLR4 system, so as to take effect on anti-inflammation and immune-regulation relevant to strengthening the body resistance and eliminating pathogens in traditional Chinese medicine.
