何首乌二苯乙烯苷预防急性酒精性肝损伤小鼠作用及其机制 被引量:11

Protection of TSG against acute alcoholic liver injury and the underlying mechanisms

摘要 目的:评价何首乌二苯乙烯苷(TSG)预防急性酒精性肝损伤的作用及其作用机制。方法:采用乙醇灌胃法复制急性酒精性肝损伤小鼠模型,通过测定血清和肝脏生化指标以及肝脏病理改变,评价何首乌TSG对急性酒精性肝损伤的预防作用,并从肝脏氧化应激方面探讨其作用机制。结果:与模型组相比,提前给予中、高剂量何首乌TSG可显著降低小鼠肝脏系数,低、中、高剂量何首乌TSG可使小鼠血清中ALT、AST活性显著降低;使肝匀浆中的TG含量显著降低,而TP含量分别提高了15.55%、20.38%和28.73%(P<0.05,P<0.01)。经透射电镜观察,提前给予何首乌TSG可改变肝细胞的超微结构,使细胞内脂滴减少,细胞核变圆变大。与模型组相比,提前给予中高剂量何首乌TSG显著降低肝脏中MDA含量;可显著拮抗酒精引起的GSH耗竭,使GSH含量恢复至正常水平;可使SOD、GSH-Px、CAT活性显著提高,且各剂量组间无显著性差异。结论:何首乌TSG可显著抑制急性酒精性引起的血清、肝脏转氨酶活性的升高,对急性酒精性肝损伤具有一定预防作用,并可能是通过促进机体抗氧化活性抑制酒精诱导的氧化应激,来发挥对酒精性肝损伤的保护作用。 Objective: To analyze the protection of TSG against acute liver injury and the underlying mechanisms exploration. Methods: The experimental model of acute alcoholic liver injury in mice was established. The serum ALT and AST activities, the ratio of liver weight to body weight, hepatie TG levels, and the hispathological changes were examined to assess the protection of TSG, and the hepatic antioxidant system were examined for its preventive mechanisms. Results: Compared with the ethanol group, the serum ALT, AST activities and the heoatic TG level in three TSG treated before ethanol groups were decreased; the TP content were increased by 15.55%, 20.38%(P<0.05) and 28.73%(P<0.01), respectively. The ultrastructure of liver cell was changed by TSG treated before enthanol under the transmission electron microscope, and the nucleus became round. the hepatic MDA content in three TSG pretreatment groups were decreased. While GSH contents were increased to the normal value. And the SOD, GSH-Px, CAT activities were increased. Conclusion: TSG could reduce the activity of serum ALT, AS, and the content TG, TP in hepatic cell, can effectively prevent acute alcoholic liver injury. TSG could significantly reduce the content of hepatic MDA, increase the GSH level and the activity of CAT. So, TSG could enhance the antioxidant capacity to cope with the oxidative stress caued by ethanol, which contributes to its protection agaist acute alcoholic liver injury.
