摘要
目的:研究青钱柳多糖对高脂血症小鼠肝脏及脂肪基因表达的影响。方法:用青钱柳多糖对高脂血症小鼠进行干预后,测定血清中TC、TG、HDL-C、LDL-C的含量,以此评定其降血脂功能;采用RT-PCR法测定肝脏和脂肪组织中PPARγ、ATGL m RNA表达水平,从分子水平探讨青钱柳多糖的降血脂机制。结果:青钱柳多糖能显著降低肝脏和脂肪组织中TC、TG、HDL-C、LDL-C的活性;同时可显著增强肝脏和脂肪组织中PPARγ、ATGL的表达量。结论:青钱柳多糖各剂量组、辛伐他汀及辛伐他汀+多糖中剂量组能够降低血清TC、TG、LDL-C值(P<0.05或P<0.01),提高HDL-C值(P<0.01);且其各剂量组均可显著或极显著提高PPARγ、ATGL m RNA表达量(P<0.05或P<0.01),表明青钱柳多糖具有显著的降血脂和提高PPARγ、ATGL m RNA表达水平的作用。
Objective: To study the hypolipemic activity and PPART, ATGL mRNA expression of Cyclocarya paliurus (Batal.) lljinskaja polysaccharide in hyperlipemia mice. Methods: The polysaccharide were fed to the hyperlipemia mice which induced with high fat emulsion for four weeks, and the TC, TG, HDL-C, LDL-C levels were detected in the liver and adipose to evaluate the hypolipemic activity; meanwhile PPART, ATGL mRNA expressions were investigated by RT-PCR in the liver and the adipose to explore the hypolipemic mechanism of the polysaccharide. Results: Cyclocarya paliurus (Batal.) lljinskaja polysaccharide could significantly decrease the TC, TG, LDL-C levels and enhance the HDL- C activities, and improve the PPART, ATGL expression levels in the liver and adipose. Conclusion: The present results showed that the TC, TG and HDL-C in the serum were significantly increased(P〈0.05 or P〈0.01), whereas the HDL-C level was decreased (P〈0.01) with the treatment of different doses polysaccharide; and a significant increase in expres- sion of PPART and ATGL could be found with different doses polysaccharide. The results indicate that the Cyclocarya paliurus (Batal.) lljinskaja polysaccharide had good hypolipemic effcects and improved the PPART, ATGL mRNA expres- sion in vivo.
