腹水传代Walker256乳腺癌细胞系建立骨癌痛模型的可行性 被引量:4

The Feasibility of Ascites Passaged Walker256 Breast Cancer Cell Lines to Establish the Model of Bone Cancer Pain

摘要 目的:探讨应用腹水传代培养Walker256大鼠乳腺癌细胞系建立wistar大鼠胫骨癌痛模型的可行性。方法:将体重180.200gWistar大鼠随机分为三组:正常对照组(Control组)、假手术组(Fake组)、接种Walker256乳腺癌细胞组(Model组)。Model组为将含1×10^8个/mLWalker256大鼠乳腺癌细胞悬液20μL注入Wistar大鼠胫骨上段骨髓腔制备的骨癌症疼痛模型。Fake组经微量进样器注射等量的生理盐水入骨髓腔;Control组则不进行手术接种,分别于手术后数天(post-cancer cell implantation day,PID)PID0d、7d、14d及21d摄片检查手术侧胫骨,观察大鼠的疼痛行为学变化,PID0d、7d及14d行胫骨HE染色。结果:PID7d摄片检查提示骨密度不均一,HE染色见大量肿瘤细胞浸润、骨小梁破坏,PID14dModel组均与Fake组、Control组行为学方面有显著的统计学差异(P〈0.01)。结论:应用腹水传代培养Walker256大鼠乳腺癌细胞系可以建立wistar大鼠胫骨癌痛模型。 Objective: To investigate The possibility of ascites cultured Walker256 rat breast cancer cell line to establish the model of tibial cancer pain in Wistar rat. Methods: Wistar rats weighing 180-200g were randomly divided into three groups: normal control group (Control group), sham group (Fake group), cancer cells inoculated group (Model group). In the model group, 20 μL Walker 256 breast cancer cell suspension with a cell density of 1×10^8/mL was injected into rat tibia bone marrow cavity. Fake animals were injected with an equal volume of saline; Control group did not undergo any surgical procedure. The X-ray examing, body weighting and pain behavior testing were carried on post-cancer cell implantation day (PID) 0, 7, 14 and 21, respectively. The tissue used for HE staining was removed on PID 0, 7 and 14. Results: In PID 7 d examination showed that the bone density was not uniform, and HE staining showed a large number of tumor cell infiltration and bone trabeeular destruction, in PID 14 d Model group, with Fake group and Control group had significant difference in pain behavior (P〈0.01). Conclusion: A wistar rat model of bone cancer pain has been successfully established by using ascites cultured Walker 256 rat breast cancer cell line.
