摘要
采用2种不同方法检测云南半细毛羊毛囊干细胞(hair follicle stem cells,HFSCs)凋亡情况,为后续的诱导分化研究奠定基础。利用凯基TUNEL-细胞凋亡原位检测试剂盒和Annexin V-EGFP/PI标记结合流式细胞仪检测HFSCs凋亡。结果:凯基TUNEL-细胞凋亡原位检测试剂盒检测表明,第2代细胞凋亡率6.0%,第6代细胞凋亡率6.5%;Annexin V-EGFP/PI标记结合流式细胞仪检测表明,第2代细胞凋亡率0.24%,第6代细胞凋亡率0.80%。说明云南半细毛羊HFSCs细胞生长状态很好。随着传代次数的增加,细胞凋亡率有所上升。
The apoptosis of hair follicle stem cells(HFSCs)in Yunnan semifine wool sheep were detected by two different methods in order to carry out the subsequent research about induction and differentiation of HFSCs. The TUNEL- Cell Apoptosis in situ Detection Kit was used to detect apoptosis of HFSCs. The combination of the Annexin V- EGFP/PI staining and flow cytometry was used to observe early apotosis occurring in HFSCs. The results from the TUNEL- Cell Apoptosis in situ Detection Kit indicated that the frequency of the apoptotic cells in the second generation was 6.0%. The frequency of the apoptotic cells in the sixthgeneration was 6.5%.The data from Annexin V- EGFP/PI staining showed that the apoptotic rate of the HFSCs in the second gerenation was0.24%;the apoptotic rate of the HFSCs in the sixth generation was 0.80%. It was concluded that the growth state of HFSCs in Yunnan semi- fine wool sheep was excellent. However, the frequency of apoptotic cells in HFSCs showed a steady increase with prolonging of the passage numbers.
