乳腺癌组织IL-17 A与VEGF表达及微血管密度临床意义研究 被引量:2

摘要 目的探讨白细胞介素-17 A(interleukin-17 A,IL-17 A)在乳腺癌发生、发展中的作用及其与VEGF、微血管密度的相关性。方法采用免疫组化法检测70例乳腺癌组织和对应癌旁正常组织中IL-17 A、VEGF蛋白和CD 34标记微血管密度表达情况。结果乳腺癌组织中IL-17 A、VEGF、CD 34阳性率分别为78.6%(55/70)、68.6%(48/70)和64.2%(45/70),显著高于癌旁正常乳腺组织24.3%(17/70)、30.0%(21/70)和31.4%(22/70),差异有统计学意义(P<0.05)。有淋巴结转移的患者IL-17 A阳性率显著高于无淋巴结转移患者(P=0.005),Ⅲ+Ⅳ期患者IL-17 A阳性率显著高于Ⅰ+Ⅱ期患者(P=0.018)。IL-17 A表达与患者年龄、肿瘤直径、组织学分级、血管神经淋巴管侵犯、PR及ER水平无关。IL-17 A与VEGF、CD 34在乳腺癌中的表达呈正相关(r=0.688,P=0.0032;r=0.724,P=0.0015)。乳腺癌组织中IL-17 A阳性表达组MVD显著高于阴性表达组(P=0.000)。结论乳腺癌组织中IL-17 A、VEGF、CD 34高表达,IL-17 A高表达可能通过促进VEGF,进而促进血管生成促进肿瘤进展,IL-17 A可望成为乳腺癌抗血管治疗的新靶点。 Objective To investigate the expression of interleukin-17 A(IL-17 A) in breast cancer and to find out the relationship between the expression of IL-17 A,VEGF and microvessel density. Methods The immunohistochemical method was used to detect the expressions of IL-17 A, VEGF protein and the microvessel density of CD 34 markers in 70 cases of breast cancer tissues and corresponding adjacent normal tissues, and their relationship with clinicopathological features were analyzed. Results The positive expression rate of IL-17 A, VEGF and CD 34 in breast cancer tissues was 78.6% (55/70), 68.6% (48/70) and 64.2% (45/70), significantly higher than that in adjacent normal tissues of 24.3% (17/70), 30.0% (21/70) and 31.4% (22/70). The positive rate of IL-17 A in cases with lymph node metastasis was significantly higher than that without lymph node metastasis (P=0.005). The positive rate of IL-17 A in re+IV stages was significantly higher than that in I + I/stages (P=-0.018). There were no relationship between the expression of IL-17 A and age, tumor diameter, histological grading, vascular nerve lymphatic invasion, PR and ER level. The expression intensity of IL-17 A was positively correlated with VEGF and CD 34 in breast cancer tissues (r=0.688, P=-0.0032 r=0.724, P=0.0015 ). MVD in positive expression of IL-17 A group was higher than that in negative group(P=0.000). Conclution The expression of IL-17 A,VEGF,CD 34 are high in breast cancer, IL-17 A might affect breast cancer by promote VEGF expression in angiogenesis, IL-17 A is expected to become a new target for anti-an^io^enic therapy in breast cancer.
