摘要 目的:研究丰城鸡血藤提取物刺芒柄花素在大鼠肠道孵育液中的代谢特征。方法:应用超高压液相色谱串联四级杆飞行时间质谱技术(UPLC/Q-TOF-MS/MS)分析刺芒柄花素的代谢产物,利用碰撞能量梯度(MSE)和质量亏损过滤(MDF)技术,并通过与文献数据或对照品比较,推测代谢产物的结构。结果:在肠道孵育液中检测到刺芒柄花素原型成分7-羟基-4'-甲氧基黄酮和13种代谢产物。结论:刺芒柄花素在大鼠肠道菌群发生广泛代谢,主要包括成羟化反应、氢化反应、甲基化和去甲基化反应以及葡萄糖醛酸化反应,为丰城鸡血藤中黄酮类成分刺芒柄花素的体内代谢特征研究提供了依据。 Objective: To explore the metabolic characteristics of formononetin in Millettia nitida Benth.var. hirsutissima Z. Wei extract when co-incubated with rat intestinal flora. Methods: An ultra-high performance liquid chromatography / quadrupole time-of-flight mass spectrometry( UPLC / Q-TOF MS / MS) method was applied to analyze the metabolites of formononetin. The structures of the metabolites were determined by using MSE and mass defect filter techniques and comparison with the literature or reference substance. Results: Formononetin prototype7-hydroxy-4'-methoxy flavone and other 13 metabolites of formononetin were detected in rat intestinal flora. Conclusion: Various types of metabolitic reactions happened to formononetin in intestinal bacteria including hydroxylation,hydrogenation,methylation,demethylation and glucuronidation,which may provide scientific basis for study on the metabolism characteristics of formononetin in Millettia nitida Benth. var. hirsutissima Z. Wei in vivo.
