家蚕雄蛾中睾酮和β-蜕皮激素的联合提取及含量测定 被引量:6

摘要 为了开发家蚕雄蛾保健食品并建立可控的原料质量标准,研究对家蚕雄蛾的睾酮和β-蜕皮激素2种活性成分进行联合提取及检测的方法。结果表明,采用70%乙醇作提取溶剂对家蚕雄蛾中的睾酮和β-蜕皮激素进行联合提取,能够有效获取2种活性成分,解决了因蛾体含有大量油脂给样品处理带来的困难,更加符合产品生产工艺要求。应用高效液相色谱法(HPLC)在检测波长为242 nm,样品溶液质量浓度20~70μg/m L的线性范围内,测定未交配家蚕雄蛾提取液中的睾酮与β-蜕皮激素的质量比分别为5.82μg/g和17.08μg/g,该检测方法的样品回收率分别达到98.59%和98.35%,可用于家蚕雄蛾原料及其提取液中2种活性成分含量的检测。 In order to develop male silkworm moths health food and establish a controllable quality standard of raw mate- rial, the combined extraction and detection methods of two kinds of active ingredients namely testosterone and β-ecdys- one in male silkworm moths were studied. The results showed that using 70% ethanol as extraction solvent can effectively extract testosterone and β-ecdysone in male silkworm moths. It resolved the difficulty in sample treatment caused by a lot of fat contained in silkworm moths, and was more in line with actual production process. Under the conditions of detection wavelength 242 nm and linearity ranges of the mass concentration of sample solution 20-70 μg/mL, the mass ratios of testosterone and β-ecdysone in un-mating male silkworm moths were determined to be 5.82 μg/g and 17.08 μg/g respectively by high-performance liquid chromatography (HPLC). The sample recovery of testosterone and β-ecdysone reached 98.59% and 98.35% respectively, indicating that this detection method can be used to detect the contents of these two kinds of active ingredients in male silkworm moth and in its extractives.
