帕瑞昔布钠对Ⅲ度烧伤早期小鼠创面愈合的影响

摘要 目的 评价帕瑞昔布钠对Ⅲ度烧伤早期小鼠创面愈合的影响.方法 雄性ICR小鼠90只,6~8周龄,体重20~25 g,采用随机数字表法,将其分为3组(n=30):烧伤组(B组)、低剂量帕瑞昔布钠组(LP组)和高剂量帕瑞昔布钠组(HP组).LP组和HP组分别于烧伤后4h腹腔注射帕瑞昔布钠0.75、7.50 mg/kg,每间隔24 h再给予1次,共给药3次,B组给予等容量生理盐水0.5 ml.每次给药后24 h(T1-3)处死小鼠10只,取烧伤创面及距创面边缘组织,进行大体观察;HE染色,光镜下观察病理学结果;采用免疫组织化学法测定血管内皮细胞标志物第Ⅷ因子相关抗原表达,进行微血管数计数.结果 与B组比较,LP组各时点时创面炎性反应无明显差异,HP组T3时创面炎性反应明显减轻,创面愈合较好.与B组比较,HP组T3时创面微血管计数升高(P<0.05),T1,2时创面微血管计数差异无统计学意义,LP组各时点创面微血管计数差异无统计学意义(P>0.05).结论 帕瑞昔布钠7.50 mg/kg可促进Ⅲ度烧伤早期小鼠创面愈合. Objective To evaluate the effect of parecoxib on wound healing in the early third degree burn in mice.Methods Ninety ICR male mice, aged 6-8 weeks, weighing 20-25 g, were randomly divided into 3 groups (n =10 each) using a random number table: burn group (B group);low-dose parecoxib group (LP group);high-dose parecoxib group (HP group).In LP and HP groups, at 4 h after Ⅲ degree burn model was established, parecoxib 0.75 mg/kg and 7.50 mg/kg were injected intraperitoneally, respectively, every 24 h (administration 3 times in total).The equal volume of normal saline 0.5 ml was given in group B.Ten rats were sacrificed at 24 h after each administration (T1-3), the tissues from and around the burn wound were obtained and observed for microscopic examination (under light microscope), and for determination of endothelial marker factor Ⅷ-related antigen expression, and microvessel count.Results Compared with group B, no significant change was found in inflammatory responses in the wound at each time point in group LP, and the inflammatory responses in the wound were significantly alleviated at T3, and the wound healing was better in group HP.Compared with group B, the microvessel count was significantly increased at T3 (P〈0.05), and no significant change was found in microvessel count at T1,2 in group HP, and no significant change was found in microvessel count at each time point in group LP (P〉0.05).Conclusion Parecoxib 7.50 mg/kg can promote wound healing in the early third degree burn in mice.
