摘要
目的观察体外微小RNA-45la(miR-451a)对胃癌细胞迁移能力以及侵袭能力的影响。方法以人胃癌细胞株BGC-823、MKN-d5为研究对象,慢病毒LV.hsa—miR-d51a感染癌细胞(Microup组),然后实时定量反转录聚合酶链反应(RT—qPCR)法检测感染细胞miR-451a基因表达水平,噻唑蓝(MTr)法检测细胞增殖能力无显著变化后,最后检测其迁移及侵袭能力:细胞迁移应用Transwell小室实验,细胞侵袭应用覆盖Matrigel基质的Transwell小室实验。设置两个阴性对照组:未感染任何病毒的癌细胞组(CON组),以及阴性对照病毒感染的细胞组(NC组)。结果比较CON组及NC组,Microup组感染细胞miR-45la基因表达水平显著增高(25.887±3.272和1.921±0.194,25.887±3.272和1.010±0.166,P〈0.05);但其迁移率降低(0.863±0.026和1.107±0.032,0.863±0.026和1.060±0.054,P〈0.05),侵袭率降低(1.040±0.055和1.522±0.001,1.040±0.055和1.696±0.078,P〈0.05)。结论miR451a在体外抑制了胃癌细胞的迁移、侵袭能力。
Objective To investigate the influence of microRNA (miR) - 451 a on migration and invasion ability of gastric cancer cells in vitro. Methods human gastric cancer cell line BGC - 823 and MKN -45 were chosen as our object. To infect the gastric cancer cells with slow virus LV - hsa - miR -451a (Micro up group), and then detect expression quantity of miR -451 a gene in the infected cells by real - time quantitative reverse transcriptase - polymerase chain reaction ( RT - qPCR). Finally, to detect the migration and invasion ability of these infected cells which were no significant change in proliferation ability by methyl thiazol tetrazolium (MTr) assay. The migration behaviors of ceils were analyzed by Transwell chamber assay, and the invasive behaviors of cells analyzed by Transwell chamber assay with coated Matri- gel. There were two negative control groups:the group of gastric cancer cells not infected by any virus (CON group), and the group of these cells infected with negative control virus (NC group). Results Comparing to CON group and NC group, expression quantity of miR- 451a gene in the infected ceils in Micro up group increased significantly (25. 887 ±3. 272 vs. 1. 921 ±0. 194, 25. 887 ±3. 272 vs. 1. 010 ± 0. 166, P 〈 0. 05 ) ; however, the migration rate (0. 863 ±0. 026 vs. 1. 107 ± 0. 032, 0. 863 ± 0. 026 vs. 1. 060 ± 0. 054, P 〈 0. 05 ) and invasion rate ( 1. 040 ± 0. 055 vs. 1. 522 ±0. 001, 1. 040 ± 0. 055 vs. 1. 696 ± 0. 078, P 〈 0. 05 ) in these cells in Micro up group all reduced. Conclusion MiR - 451 a inhibits the migration and invasion ability of gastric cancer cells in vitro. [ Key words] microRNA - 451 a; Migration ability; Invasion ability
