七福饮对糖尿病脑病模型大鼠认知障碍及神经病理改变的影响 被引量:16

The effect of Qifuyin to cognitive function and nerve pathological changes in diabetic encephalopathy rats

摘要 目的:观察七福饮对糖尿病脑病模型大鼠认知功能及神经病理改变的影响,评价其神经保护作用。方法:雄性大鼠,除空白组外均给予高脂高糖饲料,50天后腹腔注射STZ 30mg/kg建立2型糖尿病脑病大鼠模型,造模成功后,随机分为模型组、石杉碱甲组、吡拉西坦组、二甲双胍组、七福饮2.15g/kg、4.3g/kg组,给药30d后,Moriss水迷宫法进行行为学测试,结束后大鼠取血,检测血糖,部分动物处死取脑制备海马组织匀浆检测AGEs、Ach E和Ch AT,其余动物灌注取脑做病理切片,进行RAGE和NF-κB免疫组化染色。结果:七福饮可明显缩短模型大鼠逃避潜伏期、探索距离;显著延长目标象限游泳时间,增多站台穿越次数;降低空腹血糖,增加大鼠体重;升高海马组织中Ch AT活性,降低Ach E活性及AGEs水平;免疫组化染色显示海马组织DG区RAGE和NF-κB表达减少,平均灰度值增加。结论:七福饮可明显改善糖尿病脑病模型大鼠认知障碍,其机制可能是通过降血糖、改善胆碱能神经功能、改善糖尿病脑组织中AGEs-RAGE-NF-κB通路等途径实现的。 Objective: To observe the Qifuyin to cognitive function and the effect of nerve pathological change of diabetic encephalopathy model rats,evaluate its brain protection. Methods: male SD rats,except the blank group were given high fat and sugar feed,50 days after intraperitoneal injection of STZ 30 mg / kg encephalopathy in rats with type 2 diabetes model was established,after the success of the building,were randomly divided into model group,huperzine A group,Piracetam group,the metformin group,2. 15 g / kg,4. 3g / kg group of Qifuyin,after30 d,Moriss tested behavior at the water maze method,testing blood sugar,blood of rats after the end of the part of the animal to take brain hippocampus tissue homogenate detection Ach E,which and Ch AT,the rest of the animal brain tissue perfusion in brain do pathological,RAGE and nf-kappa B immunohistochemical staining. Results: Qifuyin can obviously shorten the escape latency of model rats,to explore distance( P〈0. 05 or P〈0. 01); Significantly extended target quadrant swimming time( p 0. 05),increase the platform through the number( p 0. 05); Reduce fasting blood glucose( P〈0. 05 or P〈0. 01),increase the weight of rats( P〈0. 05); Elevated Ch AT activity in the hippocampus,reduce Ach E activity and which level( P〈0. 05 or P〈0. 01); Immunohistochemical staining showed hippocampus DG RAGE and reduce the nf-kappa B expression,the average grey value increased( P〈0. 01). Conclusion: Qifuyin can significantly improve cognitive impairment,diabetic encephalopathy model rats with certain brain protection,its mechanism is through fall blood sugar,improve the function of cholinergic nerve,which improve diabetes brain tissue- RAGE- the nf-kappa B pathway and so on ways to improve diabetes mellitus model of rat hippocampal tissue lesions.
