基于人胎盘屏障体外模型对黄芩白术配伍透过胎盘药物成分的研究 被引量:4

摘要 目的:利用人胎盘屏障模型研究黄芩、黄芩白术(以下简称为芩术)配伍透过人胎盘屏障的药物成分的影响。方法:建立黄芩、芩术配伍高效液相色谱测定方法(HPLC),制备人胎盘屏障体外模型,将黄芩、芩术配伍分别加入模型母体池,在加药后10、30、60、90、120、150、180min共7个时间点采集胎儿池溶液,运用HPLC分析鉴定胎儿池溶液,研究芩术配伍对黄芩胎儿宫内暴露成分的影响。结果:实验剂量下(20mg/ml),各时间点黄芩透过胎盘的药物溶液检测的化合物中,明确归属的有黄芩苷、汉黄芩苷、黄芩素、汉黄芩素,芩术配伍透过胎盘的药物溶液检测到的化合物中,此4种化合物含量减少,以60min为例,黄芩苷、汉黄芩苷、黄芩素、汉黄芩素分别减少2.50±0.02、2.90±0.01、0.51±0.01、1.86±0.01μg/ml,具有显著性差异。结论:芩术配伍拮抗黄芩药物成分透过胎盘屏障。 Objective: Using human placental barrier in vitro model to research Skullcap,Compatibility of Skullcap and Atractylodes pharmaceutical ingredients through the placenta barrier. Methods: Establish Skullcap,Compatibility of Skullcap and Atractylodes High Performance Liquid Chromatographic method for determination. Skullcap,Compatibility of skullcap and Atractylodes were added to human placental barrier vitro model parent side,and then at 10,30,60,90,120,150,180 min obtain Fetal pool solution,using HPLC analysis ingredients of solution. Results: Under the experimental dose( 20 mg / ml),there are four compounds can be affiliation of Skullcap through human placental barrier in vitro model compounds,they are baicalin,wogonoside,baicalein,wogonin,these four kinds of compounds of Compatibility of skullcap and Atractylodes through the human placental barrier in vitro are decreased,In the case of 60 min,baicalin,wogonoside,baicalein,wogoninthe respectively reduced 2. 50 0. 02,2. 90 0. 01,0. 51 0. 01,1. 86 0. 01 ug / ml,difference was statistically significant( P〈0. 05). Conclusion: Compatibility of Skullcap and Atractylodes could antagonize skullcap compounds through the placental barrier.
