摘要
目的:观察双金痛立停胶囊对骨癌痛大鼠的作用及对肿瘤坏死因子α(TNF-α)含量的影响。方法:采用Walker 256乳腺癌细胞复制胫骨癌痛大鼠模型,选取模型复制成功的大鼠随机分为模型组、唑来膦酸(阳性药对照组)、双金痛立停胶囊19.6g,9.8g,4.9g生药/kg组,另同时设假手术组。测定各组大鼠给药前、后自由行走痛行为学评分、机械痛阈和热痛阈,末次给药后腹主动脉采血,运用酶联免疫法(ELISA)检测血清TNF-α的含量,观察骨组织的病理改变。结果:与模型组比较,双金痛立停胶囊19.6g生药/kg可明显降低骨癌痛大鼠的自由行走痛行为学评分,明显提高骨癌痛大鼠的机械痛阈值和热痛阈值,降低骨癌痛大鼠血清中TNF-α的含量,病理组织学检查表明,双金痛立停胶囊19.6g生药/kg能改善肿瘤引起的骨损伤。结论:双金痛立停胶囊对骨癌痛大鼠有镇痛作用,其机制可能与抑制肿瘤细胞分泌和释放TNF-α有关。
Objective: To study the effect of Shuangjintongliting Capsule on bone cancer pain and the content of tumor necrosis factor α( TNF-α)in rats. Methods: The model of bone cancer pain in rats was established by injecting Walker256 mammary gland carcinoma cells into tibial medullary cavity. The rats with bone cancer pain were randomly divided into five groups: model group,zoledronic acid group and Shuangjintongliting Capsule19. 6g,9. 8g,4. 9g / kg group. At the same time,other rats with sham-operation were selected to be the normal control group. The pain behavior score of walking,the mechanical withdrawal threshold and the thermal withdrawal latency of each group were measured before and after the treatment. After the last treatment,the content of TNF-α was detected by enzyme linked immunosorbent assay( ELISA) and the pathological change of bone tissue was observed. Results: Compared with the model group,Shuangjintongliting Capsule19. 6 g / kg could significantly decrease the pain behavior score( P〈0. 05),significantly increase the mechanical withdrawal threshold and the thermal withdrawal latency( P〈0. 05). It could markedly reduce the content of TNF-α( P〈0. 05). By the examination of pathology Shuangjintongliting Capsule could obviously alleviate the bone injury. Conclusion: Shuangjintongliting Capsule has analgesic effect,which is probably related to its inhibition of TNF-α.
