摘要
目的:通过肾虚质大鼠学习记忆与海马BDNF及其相关分子的表达变化,揭示肾藏志理论的物质基础与神经生物学机制。方法:采用"猫吓鼠"经典造模法造成先天不足加后天失养复合型肾虚质模型仔鼠,分为模型组、左归丸、右归丸组,空白组产自正常孕鼠。选用补肾中药进行调理,Morris水迷宫实验观察学习记忆功能变化,免疫印迹法检测肾虚质大鼠海马脑源性神经营养因子(BDNF)、酪氨酸激酶B(TrkB)、钙/钙调素依赖蛋白激酶2(CaMK2)和cAMP反应原件结合蛋白(CREB)表达。结果:Morris水迷宫实验:与空白组比较,肾虚组大鼠潜伏期、总路程以及第一次穿越目标区时间均有所增加;补肾药物干预后潜伏期、总路程以及第一次穿越目标区时间均有所降低。与空白组相比,模型组BDNF表达水平显著下降,药物干预后BDNF表达均显著高于模型组。结论:肾虚质模型大鼠学习记忆能力明显退化且海马BDNF蛋白表达下降,受体TrkB、CaMK2、CREB亦出现一定程度的降低,补肾药物可上调其表达水平从而纠正其异常变化。
Objective: Through learning and memory capacity and hippocampus BDNF expression of rats with kidney- deficiency constitution, reveal the material basis and neurobiological mechanisms of kidney deficiency constitution. Methods: By the way of "cat scare rat", built composite offspring rat models with deficiency and acquired dystrophy, then divided them into model group, ZuoGui pill group and YouGui pill group, and blank group offspring came from normal pregnant rats. After the treatment with kidney-~upplementing drugs, learning and memory capacity was detected by Morris water maze test, expressions of BDNF, TrkB, CaMK2 and CREB were detected by western blot method. Results: Morris water maze test: The incubation period, total distance and the first time through target area of model group were longer than blank group , while in ZuoGui pill and YouGui pill groups, these indexe~ were lower than model group. BDNF expression of model group more declined than blank group . After treatment with kidney supplementing drugs, BDNF expression was higher than model group . Conclusion: Rats with kidney-deficiency constitution, its learning and memory capacity and BDNF expression decline as well as TrKB, CaMK2 and CREB . Kidney supplementing drugs ZuoGui pill and YouGui pill can increase all of them thereby improve the brain function deg- radation.
