Zn^(2+)和Cu^(2+)调控的蛋氨酸硒抗自由基氧化作用研究

摘要 采用配位合成的方法合成[Zn(Met-Se)_2(H_2O)_2]·2H_2O(Zn-Se-Met)和[Cu(Met-Se)_2(H_2O)_2]·4H_2O(Cu-Se-Met)配合物,采取DPPH法、Fenton法和邻苯三酚自氧化法研究了Zn^(2+)和Cu^(2+)对Met-Se抗自由基氧化作用的影响,并采用流式细胞仪检测了对人肝癌Hep G2细胞增殖和凋亡的作用。结果显示,Zn^(2+)和Cu^(2+)均可在一定范围内提高Se-Met对特定自由基的清除率;Zn^(2+)和Cu^(2+)均可增强Se-Met对人肝癌Hep G2细胞增殖抑制作用,并明显促进该细胞凋亡,10μmol/L的Se-Met、和Cu-(Se-Met)_2诱导的Hep G2凋亡率分别为40.01%,72.84%和59.89%。 [Zn(Met-Se)2(H20)2].2H20 (Zn-Se-Met) and [Cu(Met-Se)2(H20)2]·4H20 (Cu-Se-Met) were synthesised by coordination methods. DPPH, Fenton, and pyrogallol autoxidation were used to study the anti-free radical oxidation of these complexes. The results show that Zn2+ and Cu2+ can improve free radical scavenging rate of Se-Met at a certain concentration. Zn2+ and Cu2. can enhance the Inhibition of HepG2 cell proliferation and significantly promote the cell apoptosis. Apoptosis rate of HepG2 induced by Se-Met (10 lamol/L), Zn-Se-Met (10 μmol/L) and Cu-Se-Met (10 I, unol/L) were 40.01%, 72.84 % and 59.89 % respectively.
