摘要 目的探讨乙型肝炎病毒(HBV)对胶原三股螺旋重复蛋白1(CTHRC1)表达的影响及其调节的分子机制。方法将HBV感染性克隆p HBV1.3及其单个基因的质粒分别与CTHRC1基因启动子共转染Hep G2细胞,并测定荧光素酶的活性;采用反转录PCR和Western blot检测CTHRC1 mRNA和蛋白表达的情况。结果 HBV X蛋白能够在Hep G2细胞显著激活CTHRC1基因启动子的活性,并在mRNA和蛋白水平上调CTHRC1的表达。结论 HBV通过X蛋白上调CTHRC1的表达。 Objective To explore the effects of hepatitis B virus (HBV) on the expression of collagen triple helix repeat con-taining 1 (CTHRC1) and its molecular regulatory mechanism. Methods HepG2 ceils were co- transfeeted with CI'HRC1gene promoter and HBV infectious clone pHBV1.3 and plasmids carrying individual genes of HBV, and then luciferase activitywas measured, mRNA and protein expression of CTHRC1 were measured by RT PCR and Western blot. Results HBV Xprotein could activate CTHRC1 gene promoter activity as well as up - regulate CI'HRC1 mRNA and protein expression in HepG2cells. Conclusion HBV up- regulates the expression of CHTRC1 via HBV X protein.
