摘要
目的观察麻枳降浊方干预前后便秘型肠易激综合征大鼠肠组织中5-羟色胺(5-HT)及肥大细胞(MC)水平,探讨该方治疗便秘型肠易激综合征的可能作用机制及作用靶点。方法将60只雄性Wistar大鼠随机分为正常组、模型组、阳性药物组及麻枳降浊方高、中、低剂量组,各组大鼠正常饲养,不限食水,除正常组外各组均通过冰水灌胃法建立便秘型肠易激综合征大鼠模型,模型建立后,阳性药物组给予西沙必利3.6 mg/(kg·d)灌胃,麻枳降浊方高、中、低剂量组分别给予麻枳降浊方颗粒剂25.2 g/(kg·d)、12.6 g/(kg·d)、6.3g/(kg·d)灌胃,模型组给予常温蒸馏水灌胃。肉眼观察大鼠的一般情况。灌胃14 d后处死大鼠,采用免疫组织化学法检测大鼠肠道组织中5-HT表达水平,甲苯胺蓝改良法染色后观察大鼠肠道组织内MC计数。结果与模型组相比,各给药组大鼠一般情况明显好转,肠道组织中5-HT表达水平显著下降(P<0.05),MC计数明显减少(P<0.05)。结论麻枳降浊方可能通过降低肠道组织中5-HT的表达、减少MC的数量,从而改善肠道敏感性及动力异常,起到治疗便秘型肠易激综合征的作用。
Objective It is to observe the influence of Mazhi Jiangzhuo Prescription( MZJZP) on the 5- HT and number of the mast cell in the rats with constipation predominant irritable bowel syndrome( IBS- C),and explore the possible mechanism of this method in treating IBS- C and its potential target. Methods Sixty Wistar rats were randomly divided into normal group,model group,positive drug group and MZJZP high,medium,low dose groups,normal feeding,not limited to fresh wate. Except the normal group,the rat model of IBS- C in other groups were establish by gavage with icy water for 2 weeks.Then the rats were treated with Cisapride in the positive drug group,MZJZP of 25. 2,12. 6 and 6. 3 g /( kg·d) in the MZJZP high,medium and low dose groups,and distilled wate in the model group. The general condition of rats was observed by the naked eye. After 14 days of treatment the rats were killed to observed the mast cell count in the rats of intestinal tissue by HE method,and the level of 5- HT by immunohistochemistry. Results Compared with the model group,the other rats with IBS-C were significantly improved in general,the expression of 5- HT was significantly decreased( P 〈 0. 05),and the mast cell count was markedly decrease in the intestinal tissue( P 〈 0. 05). Conclusion MZJZP maybe decreased the expression of 5-HT in the intestinal tissues,and reduce the number of MC,thereby improving the sensitivity and intestinal motility disorders to treat IBS- C.
