荞麦花叶发酵醇提物对大鼠餐后血糖的影响及机制探讨 被引量:2

摘要 目的观察麦花叶发酵醇提物(简称醇提物)对大鼠餐后血糖的影响,并探讨其机制。方法将40只大鼠随机分为低、中、高剂量组和对照组各10只,分别以30、60、120 mg/kg醇提物及纯净水灌胃,给药20 min后葡萄糖2.5 g/kg灌胃,分别于给糖0、60 min尾尖取血测血糖,计算血糖抑制率。另取20只大鼠,处死后制备小肠外翻肠囊;将18只肠囊随机分为A、B、C、D、E组和对照B组,分别置于质量浓度为0.5、1.0、2.5、12.5、25 mg/m L醇提物及生理盐水中,培养2 h后取肠囊内液体用葡萄糖试剂盒测定OD值,计算各组葡萄糖抑制率。结果各组给糖60 min血糖水平为高剂量组<中剂量组<低剂量组<对照组;低、中、高剂量组血糖抑制率分别为17%、36%、76%(P均<0.05)。A、B、C、D、E组的OD值分别为0.206±0.005、0.173±0.010、0.159±0.013、0.114±0.011、0.231±0.008,P均<0.05;葡萄糖抑制率分别为10.80%、25.11%、31.17%、50.65%、57.14%(P均<0.05)。结论醇提物可降低GK大鼠的餐后血糖,且浓度越高,作用越强,可能与其抑制肠管对葡萄糖有关。 Objective To observe the ethanol extracts from fermented leaves of buckwheat flowers ( hereinafter re-ferred to as alcohol extracts ) on postprandial blood glucose in rats and its mechanism .Methods Forty rats were randomly divided into the low-dose, medium-dose and high-dose groups and the control group A (n=10), which were respectively administered by 30, 60 and 120 mg/kg alcohol extracts and purified water .After 20 min, 2.5 g/kg glucose was used for intragastric administration .Meanwhile , the blood glucose was measured at 0 and 60 min.The inhibition rate of blood glu-cose in each group was calculated .We took another 20 rats to kill them to make the everted intestinal sacs .Then we divid-ed the 18 everted intestinal sacs into groups A , B, C, D, E and the control group B , which were respectively placed to 0.5, 1.0, 2.5, 12.5, 25 mg/mL ethanol extracts and purified water .After training for 2 h, we took the liquid within in-testinal sacs to measure the OD value with glucose reagents box and calculated the glucose inhibition rate of each group . Results The blood glucose level at 60 min: the high-dose group 〈medium-dose group 〈low-dose group 〈control group A, and the glucose inhibition rates of the high-dose group, medium-dose group and low-dose group were respectively 17%, 36%and 76%(all P〈0.05).The OD values of groups A, B, C, D, E and the control group B were 0.206 ± 0.005, 0.173 ±0.010, 0.159 ±0.013, 0.114 ±0.011 and 0.231 ±0.008 (all P〈0.05), and the glucose inhibition rates were 10.8%, 25.11%, 31.17%, 50.65%and 57.14%, respectively (all P〈0.05).Conclusions The ethanol extracts may reduce the postprandial blood glucose in GK rats , and the higher the concentration is , the stronger the effect will be, which may be achieved by inhibiting the intestinal absorption of glucose .
