摘要
以连翘的生晒果实和干燥叶片为试验材料,分别采用改良的CTAB法、高盐低p H值法和SDS法3种不同方法提取其总DNA,用琼脂糖凝胶电泳检测获得的DNA质量,并从中找出提取连翘总DNA的最适方法。结果表明,对于连翘的生晒果实,无论从产率上还是质量上,CTAB法都明显优于高盐低p H值法和SDS法;而对于连翘干燥叶片,高盐低p H值法明显优于CTAB法和SDS法。
The total DNA was isolated from the sun-dried fruit and dried leaves of Forsythia suspensa (Thunb.)Vahl by method of modified CTAB, high salt low pH method and method of SDS. The quality of the DNA was examined by the agarose gel electrophocesis. The aim was to study and find an optimum method in extracting the total DNA from Forsythia suspensa(Thunb.)Vahl. The results showed that for sun-dried Forsythia fruit, in terms of the yield or quality, CTAB method was superior to the other two methods, and for Forsythia dried leaves, high salt and low pH significantly better than the other two methods.
