摘要
目的通过检测兔皮片移植后桃红四物汤对血管生成素(angiogenin,Ang)血清含量及皮片吻合区基因表达情况的影响,探讨桃红四物汤对皮片移植中促进血管新生的作用机制。方法将75只实验兔按随机数字表法将其分为五组,即桃红四物汤低、中、高剂量组、模型组和假手术组,每组15只。造模后分组干预,并于术后2、5、8 d三个时限,从每组随机抽取5只实验兔,心脏采血后予以耳缘静脉注射空气处死,并切取移植皮片吻合区皮片组织。通过ELISA和RT-PCR检测血清样品中Ang的含量及皮片吻合区基因表达情况。结果桃红四物汤三组中移植皮片的新生血管密度明显高于模型组及假手术组(P<0.05),差异有统计学意义。Ang血清含量及基因表达中桃红四物汤三组表达量高于模型组及假手术组(P<0.05),差异有统计学意义。结论桃红四物汤能上调组织中血管新生促进因子Ang的表达,促进移植皮片成活。
Objective To investigate the mechanism of Taohongsiwu Decoction on promoting angiogenesis skin grafby detectingserum angiogenin (Ang) concentration and gene expression. Methods 75 rabbits were randomly divided into 5 groups, namely, low dose Taohongsiwu Decoction group, middle dose Taohongsiwu Decoction group, high dose Taohongsiwu Decoction group, model group and sham operation group, 15 rats in each group. After grouping intervention, at the three time-points: 2d, 5d, 8d, 5 rabbits were randomly selected from each group.After blood collection, the rats were executed by air injection of the ear vein, and the skin grafts anastomosed area skin tissues were cut. The Ang content in serum samples and gene expression in anastomosed area were detected by ELISA and RT -PCR. Results The microvessel density of skin grafts in Taohongsiwu Decoction groups was significantly better than the model group and operation group (P〈0.05), the difference was statistically significant. Ang content in serum samples and gene expression in the Taohongsiwu Decoction groups were higher than that of the model group and operation group (P〈0.05), the difference was statistically significant. Conclusion Taohongsiwu Decoction can upregulate Ang expression in the tissues to promote the survival of skin graft.
