正交法优化黑胡萝卜色素提取工艺 被引量:2

Optimization on extraction of pigment from black carrot by orthogonal test

摘要 以黑胡萝卜干粉为原料,纤维素酶溶液为提取剂,色素提取率为考察指标,通过单因素及正交试验,研究了浸提温度、浸提液pH、料液比、纤维素酶浓度和提取次数等因素对黑胡萝卜色素提取效率的影响。结果表明:浸提温度、酶浓度对色素的提取率影响较小,而浸提液pH、料液比、浸提次数对提取率的影响显著,其影响程度依次为:浸提液pH>料液比>浸提次数。同时,经过正交试验,确定了提取黑胡萝卜色素的最佳工艺条件为:浸提液pH为3.9、料液比为1:30、浸提2次、浸提温度为60℃、纤维素酶浓度为0.5%。 In order to evaluate the effects of temperature, leaching solution p H, solid-liquid ratio, enzyme concentration and times of extraction on black carrot pigment extraction efficiency, we used black carrot as raw material, cellulose enzyme solution as the extraction agent to investigate the pigment extraction efficiency through single factor and orthogonal experiments. Results showed that the effects of temperature, enzyme concentration on the pigment extraction efficiency was insignificant, and the p H, ratio of feed liquid, extraction times significant affected the extraction efficiency, and the order of the three factors affected pigment extraction efficiency from black carrot was leaching solution p Hsolid liquid ratio times of extraction. Determine the orthogonal optimization black carrot pigment extract the optimum technological conditions for leaching solution p H, material liquid ratio, times of extraction, extraction temperature, enzyme concentration was 3.9, 1:30, 2 times, 60 ℃ and 0.5%, respectively.
