摘要
目的:比较脑组织冰冻切片与石蜡切片免疫组化的效果。方法:健康雄性昆明小鼠20只,腹腔注射匹罗卡品诱导癫痫持续状态模型。24h后成功模型小鼠脑灌注固定,断头取脑,随机分别取8只行冰冻切片40μm(A组)与石蜡切片4μm(B组),采用羊抗鼠DCX(1:200)抗体,以SABC法进行免疫组化染色。结果:A组阳性神经元数目与B组比较差异有统计学意义(P<0.05),且A组镜下视野切片染色更加清晰,对比良好。结论:脑组织切片免疫组化染色采用冰冻切片漂片法效果更为可靠。
Objective:To investigate the differences of frozen sections floating and paraffin sections patching for brain tissue in immunohistochemical staining.Methods:Twenty healthy male KM mice were intraperitoneally injected pilocarpine to induce status epilepticus.24 hours after administration,the successful models were perfused and fixed,and then these brains were removed and randomly divided into two groups:Group A of 40 um frozen sections and Group B of 4μm paraffin sections.These brain sections were immunohistochemical stained with goat anti-mouse DCX(1∶ 200)antibody in SABC way.Results:The number of positive neurons was significantly different in Group A as compared with Group B(P<0.05),sections in Group A were stained more clear and contrasted better.Conclusion:The method of frozen sections floating is more reliable for brain tissue sections in immunohistochemical staining.
