大鼠骨骼肌缺血再灌注致肺损伤的实验研究 被引量:1

摘要 目的观察大鼠骨骼肌不同缺血时间再灌注造成肺损伤的变化。方法将雄性Wistar大鼠24只随机分为4组,Ⅰ组为对照组,仅进行常规麻醉,不阻断后肢血流;Ⅱ组为阻断后肢血流2 h再灌注6 h;Ⅲ组为阻断后肢血流4 h后再灌注6 h;Ⅳ组为缺血8 h后继续再灌注6 h;检测大鼠血清乳酸脱氢酶(LDH)、肺组织丙二醛(MDA)含量、肌肉组织及肺组织的病理变化。结果(1)LDH水平:实验后,Ⅰ组变化不明显,Ⅱ、Ⅲ、Ⅳ组呈上升趋势,与Ⅰ组比较,差异有统计学意义(P<0.05),且Ⅳ组>Ⅲ组>Ⅱ组(P<0.05)。(2)MDA水平:实验后,Ⅰ、Ⅱ组变化不明显,Ⅲ、Ⅳ组呈上升趋势,且Ⅳ组>Ⅲ组>Ⅱ、Ⅰ组(P<0.05)。(3)肌肉组织光镜结果随缺血时间的延长损伤依次加重,Ⅳ组骨骼肌组织细胞大片坏死。而肺组织损伤仅见在Ⅲ、Ⅳ组,且程度逐渐加重。结论大鼠骨骼肌缺血再灌注后组织细胞损伤依缺血时间的延长而加重,缺血4 h再灌注6 h时可继发肺损伤。 Objective To observe the changes of lung injury caused by different ischemia time of skeletal muscle.Methods Twenty-four male Wistar rats were randomly assigned for four groups,group I as the control group,only general anesthesia,did not block the hind limb blood flow;group Ⅱ for blocking hind limb blood flow for 2 h and reperfusion for 6 h;group Ⅲ for blocking hind limb blood flow 4 h and reperfusion 6 h;group IV for ischemia 8h and 6 h of reperfusion;detected rat's serum lactate dehydrogenase(LDH) and malondialdehyde(MDA) content,muscle tissue and lung tissue pathological changes.Results(1) LDH levels;after the experiment,changes in group I is nol obvious,group Ⅱ,Ⅲ and Ⅳ showed a rising trend,the difference were significant compared with group Ⅰ(P〈0.05),and the group Ⅳ group Ⅲ group Ⅱ(P〈0.05).(2) The level of MDA:after the experiment,group Ⅰ and group Ⅱ changes were not obvious,and group Ⅲ andⅣ showed a rising trend,and group Ⅳ group Ⅲ group Ⅱ and Ⅰ(P〈0.05).(3) The microscopic findings of the muscle tissue were increased with the increase of ischemia time,and the damage of the IV group was severe.But lung tissue injur only found in group Ⅲ and Ⅳ,and gradually increased the level of blocking time.Conclusion Rat skeletal muscle ischemia and after reperfusion tissue damage in accordance with the prolongation of ischemia lime and aggravating,ischemia for4 h and reperfusion 6 h can occurred with lung injury.
